Chicken-quail cross is a classic hybridization in fowl. but, the hybridization existed incompatibility to some extent,mounts of embryos died at the early stages of incubate.From our experiments ,we may conclude that the genes correlated sex differentiation from chicken and quail are different expressive, during the sex differention stages, lead to the death of hybrids embryos.And,we supposed that ER and its relate genes may play an important role in the deaths. On account of above,we do our experiments from three aspects :1. Observation of PGCs in early embryos of genetic hybrids of chicken-quailTo observe PGCs in early embryos of genetic hybrids of chicken-quail ,we adopt the method of histological section and PAS Specific staining. The results showed that PGCs of chick embryos is easy to be identified through PAS Specific staining,but it is difficult to identify the PGCs in the embryos of genetic hybrids of quail and chicken-quail. May be PGCs in early embryos of hybrids is the same as quails'PGCs,because PGCs in quail has little glycogen,so it is difficult to be stainned with PAS .2. The effect and different expression of Aromatase Gene in early embryos of genetic hybrids of chicken-quailThe enzyme Aromatase, that catabolizes the transformation of androgens into estrogens,is a restrict enzyme in the process.We had collected early living-embryos of 84,96,102,108,114,120,132 h at random.Adopted the method of RT-PCR, takenβ-actin as internal standard to determined the relative quantity of Aromatase mRNA on the foundation of embryos sex.Our research indicate that transcripts were detected in the embryos of female but not male embryos from 84h.At 132h,get to the Maximum.At the time of 132h,there is no female embryos nearly.Aromatase is the female special-express gene.3. Sites of ER Expression in early embryos of genetic hybrids of chicken-quailPrevious reseach in early embryos of hybrids of chicken-quail indicate that the gonad differentiation of hybrids was 66~84 h of embryos development. ER expressed abnormally and the female ER mRNA expression were significantly higher than male in the periods of gonad differentiation,with the method of RT- PCR.With Whole-mount ISH, 56,66,72,84,96 h early living-embryos be collected at random for the experiment.We found that ER was expressed around the genital ridge at the time of 66-84h ,but didn't found largely express in the time of 56 and 96h.
