In the present study,PCR-SSCP method was used to detect the SNPs of Fbxl5 and Myf6 in the core group of Jinghai Yellow Chicken.The general linear model was used to analyze its relationship with growth traits and reproductive traits,it was aimed to explore the influence of the two candidate genes on growth and reproductive traits.Additional,the expression pattern of Fbxl5 and Myf6 mRNA expression in different tissues in different growth stages of the two different species were determined by relative quantitative RT-PCR.The main results were as follows:1.The Fbxl5 was selected as the candidate gene,and the polymorphism of exon of the gene was detected.The results showed that four polymorphism sites were found,of which T14286C located in exon 3,T14291C located in intron 3,T15780C located in exon 4,G19048A located in exon 5.In addition to T14286C mutation resulted in the encoding amino acid from leucine to proline was a non-synonymous mutation.The sites did not cause the change of coding amino acids,belonging to synonym polymorphism sites.We analyzed the 4 polymorphism sites formed eight haplotype combinations in the core group of Jinghai Yellow Chicken which was in the twelfth generation by PHASE2.1.Growth traits correlation analysis showed that,haplotype combination H1H8,the dominant genotypes,were higher than other haploidtype combinations in the weight at all weeks of age,and haplotype combination H1H8 and H3H8 was significantly or extremely significant higher than other haplotype combinations(P<0.05 or P<0.01)at 0,12,14,16 weeks.Reproductive traits correlation analysis showed that mean of haplotype combination H9H11 was largest in all haplotype combinations in the respect of mean of the age of production,and was significantly greater than the H1H1,H1H3,H1H4,H1H8,H1H9 combination(P<0.01).For the body weight at first egg,the weight of haplotype combination H1H2 were largest,and haplotype combination H1H2,H1M3H H1H4,H1H8 were significantly better than H8H9,H9H9 combination(P<0.05).For the weight of the first egg,the egg weight of haplotype combination H2H4 was largest,while haplotype combination H4H9 was lowest.For the average egg weight of 300 days,the average egg weight of haplotype combination H9H11 was largest,in addition to the haplotype combination H2H4 H9H9,the rest of the haplotype combinations had the extremely significant or significant difference(P<0.05 or P<0.01);About the egg number of 300 days,the egg number of haplotype combination H1H4 was highest,and haplotype combination H1H3 and H1H4 was significantly better than H3H8 combination(P<0.01).2.The Myf6 was selected as the candidate gene,and the polymorphism of CDS region of the gene was detected.The results showed that mutation site T586C was found in exon 1,and three genotypes were formed at this site,including respectively HH,Hh and hh.The mutation led to the encoding amino acid that from alanine to aspartic acid,which was a non-synonymous mutation.We analysed the correlation between the genotypes in the exon 1 of Myf6 and growth traits,the results showed that the body weight of HH,Hh,hh genotypes in the groups at 0,2,4,6 week of age had no significant difference(P>0.05)by the SPSS software.However,Hh genotypes were significantly higher than HH genotypes(P<0.01)in 8,10,12,14 and 16 weeks of age.Reproductive traits corrrlation analysis showed that genotype hh was significantly higher than HH(P<0.05)in the weight of first egg.In addition to the age of production,three genotypes of individual had no significantly difference in the rest of the traits,but the Hh genotype was better than the other two genotypes.3.The expression profiles and expression patterns of two candidate genes in different breeds were determined.On the two chicken species,candidate genes were both highly expressed in the breast and leg muscle.The expression of Fbxl5 on the male chicken showed that:the expression of candidate gene was the highest at birth in the male and female chicken of Jinmao Yellow Chicken and male chicken of Jinghai Yellow Chicken,then continuously declined and had a little increasing next.But in the female chicken of Jinghai Yellow Chicken,expression of gene had continuously increasing and then had sharp decline.The expression of Myf6 was relatively stable,on the two chicken species,the expression level was on the upward trend and reached the highest at 16 weeks.From the expression rules of two chicken species,we could see that the expression of Myf6 in breast muscle was always higher than leg muscle in different varieties and different periods.
