| Gushi chicken is the qualified local chicken in"National resources of local bird species gene bank"in China. Taken Gushi country as the center, Gushi chicken derived through a long period of closed-proliferation and natural. It holds the characteristics of rough-resistant feeding, strong disease-resistant, and excellent performance, suitable for grazing wild backyard. Along with the implementation of the national policy of returning farmland to forests, grasslands, closing hillsides to facilitate afforestation, it is necessary to breed qualified local chicken under the conditions of undestroyed vegetation, less environmental pollution, Jieliang resistant to the rough-feeding. This study used"a rough-resistant new line feeding the breeding methods"method to breed the high-quality Gushi chicken along with the relevant research on the mechanism of their rough-resistant, the activity of crude fiber cecal cellulose in different level, different period, the performance of growing of gastrointestinal, and the location, qualitative and quantitative of normal intestinal flora, we got the physiological value of the normal flora, which enrich the content of micro-ecology, to provide the scientific evidence of resistance mechanism, to probide a theoretical basis for further screening of probiotics.Based on the sample materials of selection of Gushi chicken GM line, we chosed 246 GM chicken about 280-day-old female chickens, 50 male chickens, to boost up the level of crude fiber diet with pine needles powder. We bred F0 generation successfully. Through the research on digestive metabolism on Gushi chicken, there seems a distinct discrepancy among different individuals, which provided a better evidence on the breeding of Gushi crude feeding resistant lines. Under the same condition, we made the individuals digestion rate screening by the male to female ratio of 1:9 to conform 20 family lines to generate F1 generations.Through the study on growth performance, gastrointestinal tract development and cecum cellulose activity among different level of dietary fiber in GM line 0-8 weeks old chickens, it indicated that 4.6% high cellulose diet can boost up the height of chicken, in addition, affected the weight and length of duodenum and cecum. The cecal content in the high fiber diet group is more than control group distinctively. Male more than female, but not significantly.This study showed the research on the influence on normal intestinal flora under different fiber level mixed with pine needles addition in 0~8 weeks old Gushi chicken. We separated 8 types of normal clones from the content of four parts of Gushi chicken (Duodenum, Jejunum, Ileum and Cecum), and also with the experiment on cultivation, identification and counting. So, we got the changes on number from the normal flora, and explained the physiological value and the process of colonization of Gushi chicken gut microflora. The result showed that the number of Lactobacillus is highest (11.00±0.05), followed by Bifidobacterium(10.11±0.05) and digestion MI(9.80±0.30). The number of bacteria in cecal is higher than others (p<0.01). The total number of all intestinal bacteria in high fiber group is higher than the one in low fiber group (p<0.05), while in male is higher than in female (p<0.05). The number of bifidobacteria, lactobacillus and clostridium is significantly higher than others (p<0.01), and also took the advantage position in the intestine of 56 days old Gushi chicken. |
