The Effects Of Saccharicterpenin On The T Lymphocyte Subpopulations, The Development Of Intestine And Intestinal Mucosal Immunity In Gushi Chickens

Saccharicterpenin could promote the animal growth, improve the forage inversion rate, regulate the balance between intestinal enterobacteria, enhance the organiam immunity as animal feed additive have been showed in some researches, while the report about the influence of Saccharicterpenin in the T lymphocyte subpopulations, the development of intestine and intestinal mucosal immunity in gushi chickens is very few. So this study used the Gushi chickens as materials were invided into five treatment groups: control group (basal diets), Saccharicterpeninâ… group(300mg/kg), Saccharicterpeninâ…¡group(500mg/kg), Saccharicterpeninâ…¢group (700 mg/kg) and chlortetracycling group(200 mg/kg). we applied flow cytometry and histological techniques to research the influence of the different concentrations of Saccharicterpenin and chlortetracycling on the T lymphocyte subpopulations, the development of intestine and intestinal mucosal immunity in Gushi chickens, and to investigate whether the Saccharicterpenin and chlortetracyclingthe could influence the level and ratio of the T lymphocyte subpopulations, digestion and absorption of the small intestine, mucosal immunologic barrier.(1) The results showed the thymus index, spleen index, bursa of fabricius index in the Saccharicterpenin groups were significantly higher than the control group and chlortetracycling group(p<0.05).(2) The numbers of CD4~+, CD8~+ T lymphocyte and the values of CD4~+ /CD8~+ in the Gushi chickens in Saccharicterpeninâ…¡,â…¢group were higher compared with the control group and chlortetracycling group . The numbers of CD4~+ T lymphocyte in the Saccharicterpeninâ…¡,â…¢group were significantly higher than the control group and chlortetracycling group in the 21th day(p<0.05). The numbers of CD4~+ T lymphocyte in the Saccharicterpeninâ…¢group were significantly higher in the comparison with the control group and chlortetracycling group in the 35th and 49th day. The numbers of CD4~+, CD8~+ T lymphocyte and the values of CD4~+ /CD8~+ in all groups were decreased with the ages.(3) The length of the three sections in the small intestine in the all Saccharicterpeni groups and chlortetracycling group was shorter than the control group , but the differences amount all groups were not significan(tp>0.05). The small intestinal mucosal forms in the Saccharicterpeni groups and chlortetracycling group were more complete than the control group, while these had a little changes in all treatmental groups. The highth of villus and the V/C value in the increased by 10.75%~37.3%, 13.6%~51.05 than the control group in the 21th day, and 1.48%~20.22%,6.57%~37.89%, 6.17%~15.0%, 2.77%~30.24% in the 35th and 49th day. The highth of villus were highest in duodenum, following the jejunum, the shortest in the ileum in all groups. The V/C value was decreased from duodenum to ileu.(4) The amount of the intraepithelial lymphocytes, goblet cells and IgA~+ cells in the Saccharicterpenin groups were higher than the control group and chlortetracycling group, while the morphological change of intraepithelial lymphocytes, goblet cells in all group were not significant. The number of intraepithelial lymphocyte and lgA~+cells in experimental groups was larger than that of control group. The increment were 22.9%, 45.9%, 43.3%(p<0.05)and 58.7%, 51.6%, 54.7%, respectively. The number of goblet cells in the were increased by 27.3%,65.2% and 55.7% than the control group and chlortetracycling group. The number of the intraepithelial lymphocytes and SIgA cells were decreased from from duodenum to ileum, while the goblet cells were decreased from from duodenum to ileum.Conclusion:The results showed that Saccharicterpenin could promote the development of the immune organ, increase the number of the T lymphocyte subpopulations, improve the small intestinal mucosal structure, enhance small intestinal absorption, ameliorate intestinal mucosal immunologic barrier, strengthen small intestinal mucosal immune. Chlortetracycling could improve the development of the small intestinal mucosa, but it had a lower effect than the Saccharicterpenin in other aspects. So the Saccharicterpenin could replace the chlortetracycling as a feed additive. This will be a better economic and social benefits . The doses of Saccharicterpenin in 500 mg/kg, 700 mg/kg had a better effectiveness, the differences between them were not significant. we thought the doses of 500 mg/kg was the optimization from the economic returns.
