Studies On The Relationship Between Polymorphism Of Goat POU1F1 Gene And Slaughter Traits

The polymorphisms of POU1F1 gene were analyzed by PCR sequencing technique in two goat populations(Xiao xiang yang,Guizhou Black goat) and the effects of genotypes of POU1F1 gene loci on slaghter traits(naked body weight,dressing percentage,rib eye area,waist muscle thickness, net meat weight,net meat percentage,bone weight,flesh and blood rat) were evaluated.The objects were to explore molecular markers of slaughter traits of two goat populations,so that molecular genetic markers for efficient selection of carcass traits of goat China and construction of molecular genetic marker databases of Chinese goat provided primarily.The results were as follows.1 Exon6 and intron4 have polymorphismsPCR sequencing was applied to detect polymorphisms of POU1F1 gene locis in two goat pulations.The result showed two mutations(T-to-C and T-to-G) were found in the sixth exon region,and other two mutations(C-to-T and G-to-A) were in the fourth intintron.In the sixth exon,the T-to-C mutation was detected in the 102th nucleotide of this exon.And the T-to-G mutation was detected in the 216th nucleotide of this exon.In the fourth intron,the C-to-T was detected in the 222th nucleotide of this intron,and the G-to-A was detected in the 685th nucleotide of this intron.2 The hereditary characteristics of two goat populationsPCR sequencing was applied to detect polymorphisms of POP1F1 geng locis in two goat populations.The result showed genotypes of AA,Aa,aa,BB,bb,Bb,CC,Cc,DD and Dd were at POP1 F1 gene locus in experimented goat population.In Xiaoxiang yang population,the frequencies of gen-otype and allele for POU1F1 gene locus were 0.731,0.192,0.077/0.827,0.173,0.423,0.346,0.231/0.596,0.404,0.923,0.077/0.962,0.038,0.962,0.038/0.981,0.019 respectively;In Guizhou Black population,the frequencies of genotype and allele for POP1F1 gene locus were 0.667,0.267,0.067/0.800,0.200,0.433,0.467,0.100/0.701,0.334,0.967,0.033/0.983,0.017.0.967,0.033/0.983,0.017 respectively.Distrib-uction of the genotype and allele frequency of this locus were closely in two goat populations and genotype AA,AB,CC, DD and allele A,B,C,D were dominant.This locus was at Hardy-Weinberg equilibrium(P＞0.05).3 Relationship of POU1F1 and slaughter traits of two goat populationsIn Guizhou Black population,naked body weight,dressing percentagen,waist muscle thickness of genotype AA and aa were higher than genotype Aa significantly(P＜0.05).Rib eye area,net meat weight and net meat percentage of Aa was lower than genotype AA(P＜0.05).Naked body weight,dressing percentagen,net meat weight,net meat percentage,bone weight of genotype Bb and bb were higher than genotype BB significantly(P＜0.05).Rib eye area,waist muscle thickness of genotype Bb was higher than genotype BB significantly(P＜0.05).In Xiaoxiang yang population, dressing percentage of genotype aa and AA werehigher than genotype Aa significantly(P＜0.05). Dressing percentage of genotype Bb was higher than genotype BB and bb significantly(P＜0.05).
