| This test the white-lipped deer, Gansu wapiti, Qinghai wapiti, Tianshan wapiti, Dongbei wapiti and Tarim wapiti for the study, based on mitochondrial DNA CytB complete sequence, the combination of bioinformatics methods to detect the white-lipped deer and five subspecies of wapiti the cytochrome B gene sequence polymorphism, In order to provide scientific materials for the protection and utilizing of Chinese Cerviane breeds resource.We sequenced the mitochondrial DNA Cytb for 51deers from white-lipped deer and five subspecies of wapiti. The alignment was carried out by clustalX. Phylogenetic analysis, haplotypes and nucleotide diversity were accomplished by Dnasp4.10. MEGA4.0 was used for constructing N-J tree and ME tree. NETWORK was used for phylogenetic network constructions. According to the Genbank reference sequence (AJ000022),there were 28 polymorphic sites in 51 sequences of 1140bp long, including 10 locations of transitions and 2 transversion . 17 haplotypes were discovered in 51 sequences. The proportion is 33.3%. The highest haplotype proportion is 100% in Tianshan wapiti and the lowest is 0 in Qinghai wapiti. Gansu wapiti and Qinghai wapiti were the same DNA type. Genetic diversity of the white-lipped deer,Tianshan wapiti and Tarim wapiti were very rich;Gansu wapiti, Qinghai wapiti, Dongbei wapiti were very poor.The analysis of NJ tree and ME tree show that three obvious branches were taken on white-lipped deer and five subspecies of wapiti in Chinese . 41 sequences were downloaded from Genbank. Including 4 of Africa Red Deer (access number: AY070222,AY118198,AY244489), 2 of East Asia Red Deer ( access number : AY070224,AY2444907), 10 of West European Red Deer(access number: AY070226,AY070221,AY044859,AY244491,AF489281,AY044858,AY044860,AB021099,AY070226,AY148966),3 of Middle East Red Deer(access number:AY118199,AF489280),4 of North Asia/America(access number: AF423198,AB021096,AY044862,AF423199),5of Sika Deer(access number:AB021092~AB021094,AY035876,AB021095),3 of South Asia Red Deer(access number:AY035875,AY070223,AY044861),3 of Sambar(access number:AY035874,AF423200,AF423201),7 of Balkan Red Deer(access number: AY044857,AF423195,AF423196,AF489279,AY118197,AY070225). Therefore Chinese wapiti may have two maternal ancestral populations. It is demonstrated that Tarim wapiti has closer relationship with Western Red Deer,and the Chinese unicolor may be the ancestor of. white-lipped deer. |
