From different ecoclimate region in Gansu, 31 strains of alfalfa and three reference strains for research, routine identification and 16SrDNA sequence analysis. The results are as follows:1. Colonies formed in the composite time and strains generation time, found that: all the strains tested for the fast growing rhizobium, Sinorhizobium strains CK1.2531 (Sinorhizbium melilotii), CK1.2545 (Sinorhizbium fredii), CK1 .2547 (Sinorhizbium medicar) are reference strains.2. Results of phenotype test shown that according to the genetic distance all strains tested and reference strains were clustered into two groups (I, II) which were furthered divided into five subgroups. Group I include five subgroups, subgroups I contained 14 strains and CK1.2545 (Sinorhizbium fredii); subgroups II include five strains; subgroups III was composed of GL24,GL16 strains and reference strains of CK1.2547 (Sinorhizbium medicar); subgroups IV contained QA33B, WA62A, WA24 strains; subgroups III was composed of seven strains and reference strains CK1.2531 (Sinorhizbium melilotii). Through the form of cluster figure, select of four strains by 16 SrDNA sequences comparison and phylogenetic analysis, results showed that: four strains can be divided into two species, QA46A, WA24 and WL47 is Sinorhizobium , GA26 is Rhizobium or its variant strains, the results and the results of cluster maintain a high consistency.4. Through the community morphological, physiological and biochemical of determination and 16 SrDNA sequence analysis, results showed that GA66, QA46A, WL53, QA50A, DA99, GL21, DL81, QL36B, TL18, GL65, DL67, TA34, DL15, QL31B, and CK1.2545 is Sinorhizbium fredii; DL58 , TL22A, TL47, WA32, DA10 is Sinorhizobium xinjiangense; GL24, GL16 and CK1.2547 (Sinorhizbium medicar) is Sinorhizbium medicar; according to their genetic distance, Sinorhizbium melilotii including the GA28, QL20B, DA42, DA53, WL47, WL68, QA33B, WA62A and WA24; GA26 is Rhizobium species or its variant strains.
