20 cultivars of water dropwort were selected in this study,38 morphological traits estimated.Genetic relationship of 20 varietics were evaluated by RAPD analysis.The consistency of genetic and morphological diversity among the varietics was discussed accordingly in order to establish the foundation for water dropwort genetie improvement.The main results were summarized as follows:1.The optimized RAPD to evaluate genetic diversity reaetion system was obtained,which can be used in water dropwort germplasm.This research use 20ul response system in which contains 20ngDNA,Mg2+2ul,dNTP 0.8ul,0.4ul random primer,Taqpolymerase 0.2ul,10*buffer 2ul.The temperature profile used for PCR was 94℃for min,followed by 40 cyeles of 94℃for lmin,38℃for 1min,72℃for 2min,and was terminated with a 10min DNA extension step at 72℃.2 A total of 70 10-mer random primers was used in this Study .Among them,11 Primers Produeed 117 scorable and repeatable RAPD bands,of which 113 bnads (97%) were polymoprhic across 20 water dropwort accessions.It proved that the level of polymorphism was high in 20 water dropwort germplasm.3 The cluster analysis of the genetic distance of agronomic characters showed that the cluster analysis could reflect geographical distribution of the water dropwort in some degree.The result explained geographical elements can affect varieties greatly.But the genetic distance of few materials from different regions was very simila,which might have a relation with similar natural environment and closely artificial choice of the materials.4.The cluster results according to genetic similarity coefficient and genetic distance were dieffrent. Polymoprhism detected by RAPD was the variation of DNA, But the Phenotypic was the result of genetic,enviromnental and GE interactional effect.Each mehtod can provide useful infomation.Therefore,genetic diversity of gennPlasm can bebetter evaluated by dieffrent measures so that gemrplasm can be protected and used effectively.
