Study On In-vitro Propagation Technology Of Several Oriental Lily (Lilium Oriental Hybrid)

The oriental lily(Lilium oriental hybrid) squamas of "Sorbonne"and"Acapulco",and the flower organs(petals,filaments and orary) of "Huangshidai"and "Marcoplolon",and the leaves as explants were used to establish the in vitropropagation. The four technique systems in regeneration clones,the proliferation,rooting culture and bulblet formation and transplant were studied,the results was asfollows:1,the establishment of clonesthe proliferation speed and differentiation quantity of bulb scale from the "Acapulco"and "Sorbonne" were as follows: "Acapulco">"Sorbonne",and the sequence of bulbscale was basic part >middle part > top part. The most suitable media in vitropropagation of the two varieties was MS+6-BA 2.0 mg/L+NAA 0.1 mg/L.the basic part of the scale was the most favored part for the differentiation in theinduction,and on the media of MS+6-BA 3.0 mg/L+NAA 0.3 mg/L.The differentiation ability of flower organ in"Acapulco" and " Huanshidai" was different,the speed in "Acapulco"was faster than that in "Huangshidai",and theorary was the first in differentiation of flower organ,petals was the second,and then filaments followed. The most suitable explants for multiple orary differentiation in vitro propagation was on the media of 6-BA1.0mg/L+NAA0.2 mg/L,the orary and petals and the flower of the "Acapulco" was induced to differentiate on the media of 6-BA2.0mg/L+NAA0.2 mg/L.2,proliferationThe media for proliferation of bulb scale of "Sorbonne" and "Acapulco" was on the MS+0.4 mg/L 6-BA+0.05NAA mg/L in the induction of adventitious bud,and the proliferation induction of flower organ in Huangshidai was on the media of MS+0.3 mg/L 6-BA+0.02NAA mg/L.3,rooting culture and bulblet formation when the practice and the economic factors were concerned, the most proper media inrooting culture and bulblet formation of "Marcoplolon" was l/2MS+sugar60g/L+NAA0.5 mg/L, and that of "Acapulco" and "Sorbonne" was l/2MS+sugar70g/L+NAA0.3 mg/L.4,transplantBulblet formation was the key step in the survival of transplants, Heel-in forhardening can helpto the survival rate.
