Some natural medicinal plants are in short supply because of the destruction of natural environment and unplanted exploiting.It is an effective way to get medicament through manual planting,though with many problems,such as poor quality and yield because of medicinal plants being infected by viruses,etc.So we want to find a new way of manual culture,in order to get the high quality medicinal plants.Radix Astragali is the root of Astragalus membranaceus Bge. var.mongholicus(Bge.) Hsiao and A.membranaceus.Radix Astragali is an important plant for medicine as well as popular herbal medicine in China,and AstragalosideⅣis the major secondary metobalite.We select the high quality Astragalus membranaceus to research.Research on the seed germination by different pretreatment.Using the Radix Astragali plantlets directly collected from the fields and plantlets planted in laboratory as experiment material,the induction was studied systematically from the following aspects:type of explant,size of explant,at the same time callus culture system was established. Seed,callus and young seedlings were irradiated by 60Co-γray,while the half lethal dose of them were found;at the last,Radix Astragali was mass-produced by rapid propagation.The result indicated:0.5 mg/L GA3 was the best effective on seed of Radix Astragali germination in one year and it could percentage increasing 20%, 0.8 mg/L GA3 was the best effective on seed of two years,and it was same as the one year seeds;the callus induction could be effected by the type of explant,size of explant,by optimizing the culture conditions,we attain the optimal conditions for the growth of callus was MS combined 2,4-D 1.0 mg/L,KT 0.2 mg/L and 6-BA 0.5 mg/L.The best explant is root segment.The optimal measure of the explant was that 1.0 cm root segment placed in the medium.For setting up the technique of irradiation breeding and providing high quality seedlings or seeds of Radix Astragali,the seeds,callus and seedlings of R.Astragali were irradiated by 60Co-rγay,The effects of irradiated seeds was better than irradiated callus and seedlings.So the seeds was the best to effects of irradiation in three materials,and the contents of AstragalosideⅣin roots of plantlets could be increased by irradiated seeds. in the end,irradiated the seed by 400 Gy is the best way of irradiation breeding.AstragalosideⅣwas extracted and determinated by HPLC.The contents of AtragalosideⅣreached to 0.25 mg/g in roots of plantlets from irradiated seeds of R.Astragali;For select and develop the excellent, mass-produce high quality Radix Astragali seedlings and seeds,rapid propagation was done,the optimum medium for bud propagation was MS supplemented with 2.0 mg/L 2,4-D,0.5 mg/L 6-BA and 0.1 mg/L KT.The optimum medium for root induction of test-tube plantlets was MS added with 0.5 mg/L NAA and 1.0 mg/L DSC.
