Belonging to Bupleurum of Umnelliferae, Bupleurum is an important plant for medicine as well as of popular herbal medicine in China, Corea and Japan. According to Chinese Medicine Dictionary, Bupleurum Chinese DC.and B.scorzonerifolium Wild. are the two original plants for medical use. The study show: Bupleurum Chinese DC. and B.scorzonerifolium Wild. are both contain many effective chemical constitutions, so both of them are considered typical species. The Bupleurum Chinese DC. is widely found in Northern China, with its good quality. But due to the heavy demand and scarce resource, currently only 50 % of demand can be met and the other 50 % needs to be supplemented by domestic production. So the people of this region are developing domestic production, which has been developed considerably in Shanxi, Hebei, Shanxi and Gansu. But because the appearance of the different species is similar and they are often mixed up in one region, it is always very difficult to divide them from morphological characters, causing disputes on classification. After investigation, the regions which produce Bupleurum Chinese DC. have mixed up their species, with different variants in the same species, closely related species in their morphology and different paramorphia species after cultured, which have led to the mixing up of species poor quality and further medicine's inaccuracy and effect's inability, affecting the fame of exports.This experiment tries to assess and analyze the seed viability and germination rate of B. scorzonerifolium Wild., which are stored for different periods of time, to conclude the favorable conditions for the seed germination. With the Bupleurum Chinese DC. and B.falcatum directly collected from the fields, as experiment material, the author of this thesis tries to carry out culture of stem, leaf, petiole and flower bud. The author also researches the construction of rapid propagation and the process of organogenesis, discusses the exogenous hormone's function in the callus shaping process and rapid propagation of the plants, and with the paraffin method, the author alsomakes an observation and comparison of the microscope structure of Bupleurum Chinese DC. from different places.The results reveal that TTC seed viability rapid assessment method is not only rapid but also effective. The favorable germination temperature of B. scorzonerifolium Wild, used to be 20 °C, with a germination rate of 40.2 %. But after being soaked in 3 % H2O2, its germination rate rises to 52.5 %. If keep one more year, the seeds lose their germination viability. In analyzing the leaf and stem callus of Bupleurum Chinese DC, stem and bud flower are the suitable materials, which make the callus intense, with more differentiation ability. The callus stimulated by the leaf is lose and seriously browning, basically without differentiation ability. In the bud rapid propagation, 6HBA is the main hormone and the basic medium of B5 is more superior to MS. With the help of casein hydrolyzed, B5 accelerates the plants' growth;The author also observes the slides of roots and leaves of Bupleurum Chinese DC. from five different places, and finds plant from Gansu Province has lower degree of lignification than that from Shanxi Province, while plant from Longxi, Gansu has also lower degree of lignification than that from Li County Gansu Province ,which proves that the quality of the plants has something to do with its region and environment. The leaf structures of the plants from five places do not differ remarkably, with only some differences in size.
