Sodium new houtluyfonate (SNH) is the additional compound of sodium bisulfite and houttuyninum, which is the primary antibiotic ingredient in the volatile oil of the medicinal plant houttuynia cordata thunb (HCT). SNH is obtained by removed the component which is harmful or useless from HCT and the performance is stable and safe. The chickens were immunized with the LaSota live vaccine against Newcastle disease virus (NDV) in the research, to investigate the effects of SNH on chicken immune response and their optimistic dosage:Effects of SNH on proliferation in peripheral lymphocyte of chicken in vitro Anticoagulant blood were sampled from the chicken, then the peripheral lymphocytes were collected and cultured for 24h. Different dose of SNH and/or ConA were added in the lymphocytes in vitro, and cultured for 44h. The lymphocytes proliferation was determined by MTT method, and the data show that SNH could induce lymphocyte proliferative response singly or cocultivated with ConA. In addition, the effect of different dose of SNH were dissimilar on improve lymphocyte proliferation, while the optimistic concentration is 250mg/L.Effects of SNH on chicken humoral immunity To determine the effects of SNH on chicken humoral immunity, 288 healthy eleven-day-old chickens were assigned to eight groups. The first three groups were treated with different dose of SNH respectively before the vaccination, and the second three groups were treated with different dose of SNH respectively on the vaccination, the 7th group was treated with Interleukin-2(IL-2), while the 8th group only injected with normal saline water as control. All the chickens were then immunized with the LaSota live vaccine against NDV for two times with 14 days interval. On the 7th, 14th, 21st, 28th, 35th and 42nd day after the first immunization, the titer of antibody against NDV were determined, and the data show that SNH could increase antibody titer agaist NDV. In addition, the effect of different levels of SNH were dissimilar on improve humoral immunity, while the optimistic level is 2mg/kg.Effects of SNH on chicken cellular immunity To determine the effects of SNH on chicken cellular immunity, 288 healthy eleven-day-old chickens were assigned to eight groups. The first three groups were treated with different dose of SNH respectively before the vaccination, and the second three groups were treated with different dose of SNH respectively on the vaccination, the 7th group was treated with IL-2, while the 8th group only injected with normal saline water as control. All the chickens were then immunized with the LaSota live vaccine against NDV for two times with 14 days interval. On the 7th, 14th, 21st, 28th, 35th and 42nd day after the first immunization, the ratio of CD4+/CD8+ peripheral lymphocyte were determined, and the data show that SNH could improve the ratio of CD4+/CD8+ peripheral lymphocyte. In addition, The results show the same the dose-effect with experimention of Chapter three.Effects of SNH on chicken immune response in immunosuppression To determine the effects of SNH on chicken immune response in immunosuppression, 120 healthy seven-day-old chickens were assigned to 5 groups. Florfenic and three different dose of SNH were treated in the first three groups respectively, while the 4th group was orally administered with florfenic and the 5th group was only injected with normal saline water as control. All the chickens were vaccinated with the LaSota live vaccine against NDV for two times with 14 days interval. On the 7th, 14th, 21st and 28th day after the first immunization, the titer of antibody against NDV and the ratio of CD4+/CD8+ peripheral lymphocyte were determined. The data show that the oral dose of 1.2g/kg of florfenic could significantly inhibite immune response. In addition, the data revealed that SNH could depress the immunosuppression of Florfenicol on chicken.
