| Growth hormon(eGH)is one of the important factors in regulating many physiology and metabolic processes in animal body. So we detected the GH gene mutation in these 4 population such as Small-tailed Han sheep,Genmenmarino sheep ,Mgoulion sheep and hybrid generations of Genmenmarino and Small-tailed Han sheep by PCR-SSCP and DNA sequence analysis technology. The results was as following:1.By 5 primer, their were five locus were detected , which were the 1st intron ,the 2nd exon , the 2nd intron, the 3rd exon , the 4th exon , the 4th exon ,the 5th exon, in which , the 4th exon was the polymorphic Loci.2.The analysis DNA sequence showed that both the mutation of G→A in 1257bp and T→C in 1405 bp in the 4th exon took place,which resulted in the GH allelic gene mutate from A to B.3.Both the gene frequency and genotype frequency were calculated for the polymorphic Loci. The results were as following,:the frequency of A,B,AA,BB,AB, were 0.4858,0.5142, 0.2286,0.2571,0.5143 respectively inMongolian population; which were 0.3282,0.6719, 0.2188,.05625,0.2188 in Small-tailed Han sheep, 0.6055,0.3950, 0.4737,0.2632,0.2636 in Germanmerino , 0.5750,0.4113,0.4583,0.2947,0.2333 in hybrid generation 1 of Mongolian and the Small-tailed Han sheep and Germanmerino respectively. Moreover, the allelic gene A, B frequency inSmall-tailed Han sheep,Germanmerino,hybrid generation 1 of the Small-tailed Han sheep and Germanmerino were fit for equilibrium of Hardy-weinberg.4.The gene and genotype frequency analysis also showed that Germanmerino and hybrid generation 1 of the Small-tailed Han sheep and Germanmerino which have high mutton yield had the dominant genotype AA ,whileSmall-tailed Han sheepwhich have low mutton yield had the dominant genotype BB,.So we concluded that GH-exon4 might relate to the mutton yield of sheep.5.The polymorphisms analysis showed that Polymorphism information content(PIC)was medium, which from higher to lower wasMongolian(0.3748),Small-tailed Han sheep(0.3436),Germanmerino (0.3629),hybrid generation 1 of the Small-tailed Han sheep and Germanmerino (0.3883).The hoterozygosity (Ho)from higher to lower wasSmall-tailed Han sheep(0.5004), Germanmerino (0.5591), Mongolian (0.5226), hybrid generation 1 of the Small-tailed Han sheep and Germanmerino (0.4998). The heterozygosity (He) from higher to lower was hybrid generation 1 of the Small-tailed Han sheep and Germanmerino (0.4996), Mongolian (0.4409), Germanmerino (0.4774),Small-tailed Han sheep(0.5002). |
