Hericium erinaceus is a kind of precious fungus which was used for medicine and edible. The effective component of polysaccharides was widely used to exploy the enteron ulcer drugs and anticancer drugs which has been gained well Clinical effect. But the research on the metabolism rule of Hericium erinaceus in liquid fermentation needed deeply improvement. At present, the research on difference of Hercium was not yet report in China or abroad. Furthermore, the difference of species in genus is rare reported too. So we compared the metabolism production in broth, using the cultural methods of liquid shake flask and submerged fermentation in order to protuct the independence intellectual property rights.The first part, using the methods of biology and molecular biology compare Hericium erinaceus mycelium to Hericium caput-medusae mycelium. The result of rivalry experimentation, fermentation mycelial morphous, fruiting body morphous and RAMP molecule marker are different. So the two species belong to different Hericium genus.The second part, at the same condition of liquid shaking culture, put the two species in corn steep liquor medium and brown sugar medium cultivated for one month. During the cultivation, the samples of the broth were taken every two days. The experiment will use phenol-H2SO4 method,DNS method to survey the content of total sugar and reducing sugar in different samples. Also use the Coomassie brilliant blue method to detect the changes of the content of the protein in the broth. Using ultraviolet spectra scanning and fluorescence spectrum scanning detect the dynamic state change from product of metabolism. The result indicated that the changing tendency on content of sugar and protein are different ether the two strains cultivated in the same medium or in different midium. Brown sugar medium is good for the production of polysaccharides, while corn steep liquor medium is good for the production of micromolecule.The third part, Hericium erinaceus and Hericium caput-medusae were using the same condition to liquid submerged fermentation. Then choose the drug active component—polysaccharides as destination, using same method to extraction, abstraction and purification. Using 80℃water to extraction, triple 95% ethanol to precipitate polysaccharides, Sevag method to remove the nomadic protein, dialysis to eliminate the micromolecule, ethanol to fractional precipitation, DEAE cellulose column chromatography to remove some pigment and protein, Sepharose CL-6B gel column chromatography purify uniform polysaccharides. The HCP and HEP are uniform polysaccharides throth the methods of film electrophoresis and Sepharose CL-6B gel column chromatography. The molecular of HCP and HEP are 46386 and 26671. The further GC analysis proved that there are significant difference between Hericium erinaceus and Hericium caput-medusae .The composition of monosaccharide of Hericium erinaceus is Glu :Ara=1.09:1, The composition of monosaccharide of Hericium caput-medusae is Glu.According to the product of metabolismthe dynamic state and the composition of broth polysaccharides find that the two species surely exist clearly difference.
