According to literature, Hericium erinaceus contains various active substances and lots of nutrients. It is able to repair our erinaceus in tragacanth and cultivate.Along with the very fast development of edible fungus industry, the identification of new edible fungus has become a very important issue in edible fungus'breeding. DNA marker is a original research direction at home and abroad in identifying the same fungus which have new breeds and different names in recent years. DNA fingerprinting is a stable, simple and fast technique, and it can protect the right of new strains, which can solve current dispute about stains with different names or new breeds in planting fungus process.This research evaluated the strains identification of Hericium erinaceus through antagonism analysis and Inter-simple sequence rcpeat(ISSR) analysis on totally 54 cultivated strains and 11 wild strains,the selected strains were further analysed by ITS sequeusing and physiological properties.The results obtained are summarized as the following.1. Antagonism analysis was implemented for 54 cultivated strains and 11 wild strains from the plateau of west Sichuan.The morphological and the antagonistic characters indcated the plylogenetic relationship among the different Hericium erinaceus, according to width and colour of rivalry linear. The results showed that antagonism analysis could differentiate the strains,but easy to make some subjective wrong judgment.2.The genetic diversity of the 65 strains tested was evaluated using technique of Inter-simple sequence repeat (ISSR) with 8 primers.The dendrogram was construded using NTSYSpc2.1 softwore. The results of ISSR showed that the 8 selected ISSR primers could distinguish all isolates from one another, which demonstrated the high genetic diversity. The similarity coefficient of variation ranged from 0.444-0.950, and the 65 isolated were clustered into 4 groups at the similarity coefficient 0.675. It mean that the strains possess comparatively significant genetic variations.lt provides great meanings on reservation, culture and application..3.The ITS sequences of the selected strains were amplified with ITS4-ITS5 and compared with the known species of mushrooms.The sequences were compared with the data from the GenBank.The of phylogenetic tree was constructed The ITS4-ITS5 sequences of these strains were highly simllar with the type strain EU784264.1 Hericium erinaceum and AY534589.1.The tested strains were identified as Hericium erinaceus. 4. The agronomic traits including morphological features and yield of the 54 cultivated strains and 11 wild strains were tested.The results showed that:all of cultivated Hericium erinaceus strains could fruit, All of wild Hericium erinaceus strains could fruit with low yield. The yield ofstrain 63 of wild Hericium erinaceus was 160.53 g,which was higher than that of some cultivated Hericium erinaceus strains.5. This research evaluated the resource of Hericium erinaceus by determining the mycelial physiological traits of part of cultivated strains and wild strains(including mycelial growth rate on PDA, mycelial growth rate under different temperature).The result revealed that all strains stopped growing at 5℃.At 40℃,all strains stopped growing; 25℃was the most suitable temperature for the growth of Hericium erinaceus mycelium.
