This research used Narcissus tazetta L. var. Chinensis Roem as materials,and researched the change rules of Carbohydrate,soluble Proteins,POD activities,Isoenzyme,Endogenous Hormone and Polyamine changes in the narcissus bulb treated with different low temperatures,to reveal the mechanism of delaying narcissus ball aging and extending the storage time,to provide theoretical guidance for the anniversary of flowering regulation technology.The conclusions of this experiment were as follows:1.Through different low temperatures 7℃, 4℃, 1℃and CKof treatment,measured some related physiological and biochemical indexes,and watered curing in batches.By data analysis,it was found that narcissus blossom before 182 d but could not flower after 182 d under the storage of 4℃.It may be that bulbs consume more nutrients by the respiratory consumption with the storage temperature and time prolongation,so as to the nutrient storaged in bulb can not meet narcissus requirements for growth and development.So how to curb nutrient loss during cold storage need to be further studied.2. Changes of the soluble total soluble sugar, sucrose, starch and amylase activities in Narcissus were studied.The results show that: The content of total soluble sugar and sucrose increased in the storage; There was significantly negative correlation between the Starch content and amylase activities(r = 0.46279**).Low temperature promoted the starch into soluble sugar.It had appeared the phenomenon of"Low Temperature Sweetening".Levels of soluble sugar continue on the rise before the storage of 152 d, which indicated carbohydrate materials were the basis of energy and material to flower.3.Changes of the soluble protein,SDS-PAGE of soluble protein,POD activities and Isoenzyme in narcissus were studied.The results show that: Protein degradation may be necessary for the flowering mechanisms with the role of low-temperatur;The protein contents of different parts in narcissus bulbs were the highest under 4℃treatment, indicating that 4℃was better storage temperature to delay flower; Found that cold treatment increased a number of multi-band in protein electrophoretogram spectrum,which illustrated that treatment promoted new protein synthesis; There is a certain difference between different temperature and dprocessing time of POD isozymes, POD isozyme bands intensity strength were in line with POD activities,which showed POD activities decreasing with the extension of storage time.4.Changes of the Endogenous Hormones and Endogenous Polyamines in narcissus were studied. The results show that: the content of IAA sharp decline firstly and then picked up throughout the storage period, thus IAA may play a role in promoting and suppressing flower; he content of ABA run up and then declined before storage of 152 d. The content was very low, the maximum was only 0.34 ng / g.FW,ABA may break dormancy of narcissus ball; The content of GA3 was high and later took on decrease trend in early stage. High content of GA3 induced floral induction,GA3 may promote the formation of a-amylase so as to promote the formation of the hydrolysis of starch,which may inhibit the growth of the narcissus ballflowering induced; The content of ZR had a cumulative process and then showed gradual increase trend,which showed high content of ZR played a significant role in flowering induced.In the three Endogenous polyamines the content of Spm had the biggest change range, about 88% of the total polyamine.The effects of low temperature may carried out by influencing the content of Spm in the proportion of polyamine.Polyamines and Hormones can be produced in living cells, which played role in the growth and development and aging regulated.5.This research determined the measurement condition of the content that IAA, ABA ,GA3,CTKs and PAs by HPLC.
