Study On Preparation Of Propolis Liposome And Its Hypoglycemic Effect

The mainly medicine ingredient of propolis is the unstable flavonoid,which shows widespread biological activities,including oxidation resistance,anti-tumor,enhancement of immunity.However,the bitter taste and the unstable character limite the application of propolis in medicine.The liposome,a new medicine vector,has been widely used in medicine for the characters of directly target,high effective,slow release and lower toxicity. In present study,the processing parameters of propolis liposome were optimized.The physical and chemical characters,the hypoglycemic effect of propolis Iiposome were analysised.The main results are listed as followes:Through thin film supersonic method prepare propolis liposomes and its optimum parameters were phospholipids to propolis 10,phospholipids to cholesterol 3,rotating evaporation temperature 40℃,surfactant to phospholipids 0.4.Under this condition, propolis liposome encapsulation efficiency rate of 90.69%,K_E was 9.7%.The physical and chemical properties of liposome suspension and the freeze-dried liposomes were studied,including the size distribution,encapsulation efficiency,K_E,and pH,indicating that after the freeze-dried liposomes to maintain the basic physical nature of the original.Stability of results showed that the liposome freeze-dried state to improve the stability of liposomes.Hypoglycemic effects of propolis liposomes1) After administration on normal mice for 4 successive weeks,the tablet(low,moderate, high dosage) had no significant effect on these mice's weight,serum blood sugar and serum insulin,while administration for 4 successive weeks it can enhance liver glycogen content and improve glucose tolerance ability of normal mice quickly.2) Compared with the model group,4 weeks of successive administration of the tablet can significantly increase body weight and antioxidant capacity of mice,decrease the serum blood sugar,improve glucose tolerance ability,enhance liver glycogen content and improve serum insulin;base on the same effective medicine,the effect of propolis liposomes better than normal propolis.3) Compared with the model group,the pancreatic Bcl-2/Bax mRNA ratio was increased with high dosage of propolis liposomes,after administration for 4 successive weeks, effective suppression of apoptosis;base on the same effective medicine,the effect of propolis liposomes better than normal propolis..
