The pigs'Ghrelin gene plasmid were injected into piglets in the study,the purpose is to shudy it's effect to promote growth and hormone level of blood,for settling the application foundation in the cultivation.The study use the simple factor experiment,a devil of the PCI-Ghrelin-28aa gene plasmid was got out and depurated,adoption eight weaning piglets of"Junmu No.1",its' mean weight is 16.33±1.77kg,its were selected anddivided two groups,there are the experiment group and control group,each group had four repeat,each repeat had one piglet.The piglets of the experiment group.The PCI-Ghrelin-28aa gene plasmid and Elution Buffer were injected to the piglets semitendinous muscle of the experiment group in 0 day,the Elution Buffer also were injected to the piglets of the control group,the all experimental stage had 63 days.During the all experimental stage we must to record the information of weigh and foraging,on the 32,33,34,46,47,48day,we order to collect piglets' egesta,adopt AIA method to determin the CP digestibility of forage and egesta.The nine in the 0 day(befor the injecting),7,21,35,49,63 day morning,every the piglets were benificed blood 20ml from vena cava anterior,the blood sample to be divided to two share.the t blood serum to be determined:the total protein,albumin,T-CHO and growth hormone,IGF-I,growth hormone release-inhibiting hormone,gastrin.The result of the effect of PCI-Ghrelin-28aa gene plasmid On performance trait in piglets showed that:The body weight from 16kg to 60kg:injecting of PCI-Ghrelin-28aa gene plasmid increased the end weight by 3.5%,to observe all times,the average intake of all two groups are almost to equal,the average gain and the feed consumption/gain were all significant deviation(P <0.05)。From 21to 35 days the average gain of the experiment group increased by 10.76%, significant deviation(P<0.05),the feed consumption/gain dropped by 8.78%。From 49 to 63 days the average intake and the average gain dropped down than the control group,the average intake by 7.35%,the average gain by 4.73%(P<0.05),the CP digestibility increased by 7.99%(P<0.01)of the experiment group in the first stage,the control group didn't marked change,and also didn't havesignificant deviation.The result of the hormone level and the biochemical indicator determination:injecting of PCI-Ghrelin-28aa gene plasmid increased the GH level by 11.20%(P<0.05),the IGF-I level had increased tendency on the order 35day.The GAS level had increased dramatically(P<0(?)01) than the order 0,7,21,63d,and increased by 26.46%(P<0.05) than the control group.The SS level of the experiment group increased by 29.26%(P<0.05),29.17%(P<0.05)和38.17%(P<0.01) on the order 7,21,63d.The total serum protein and albumin level increased by 20.35%,29.29%(P<0.05)on the order 35day.The total cholesterol level of each group rised tendency,the oeder 35 day the total cholesterol level of the experiment group increased obviously the order 0,7,21d,on the order 49,63d the total cholesterol level increased obviously the order 0,7d,and on the order 35d the total cholesterol level increased by 9.23%(P>0.05).The applying result showed that:(1) injecting of PCI-Ghrelin-28aa gene plasmid to possessed growth promotion in the pigs,the effect time to expressed one month at least,to possessed stage of incresing CP digestibility,dropping the feed consumption/gain,and enhanced the pig immunity.(2) injecting of PCI-Ghrelin-28aa gene plasmid increased apparente the GH,IGF-I level,the high GH level to promoted gastric acid and GAS secreted on the order 35d,and at the same time also made the SS level to increased,the high SS level also suppressed the GH,IGF-I.Evidenced initial the Ghrelin gene plasmid may be applied in future.
