In this experiment, PCR-SSCP, PCR-RFLP and DNA sequencing techniques were applied to analyze the genetic variation of MSTN gene IntronⅡ, GH gene exonⅠand exonⅡtwo candidate genes three genetic loci. The objective was to detect the hereditary characteristics and its associations with some growth traits(body weight, body length, withers height, heart girth and cannon circumference). And analysis the relationship between BMP15 gene intronⅡand reproductive traits(litter size).To explore molecular markers with significant effects on important economic traits for efficient selection, and to provide genetic information for construction of molecular marker database, protection and utilization of breed resource of goat. The results were as follows:1 Associations of polymorphisms of Haimen goat MSTN gene IntronⅡand growth traitsThere were AA, AB two genotypes in MSTN gene IntronⅡ, the results indicated that AB and AA of MSTN gene's two genotypes has a mutation T→C in 3783th base. The A allele was dominated in this loci, and the group was in Hardy-Weinberg extreme imbalance(P<0.001).There was extremely significant difference between the genotypes and birth weight, AA﹥AB (P<0.01), and significant difference between the genotypes and withers height of age(P<0.05), AA>AB. There were no significant effect in 2-month weight, six-month weight and age weight, but the trend of AA>AB; for other growth traits were no significant differences (P>0.05).2 Associations of polymorphisms of Haimen goat GH gene exonⅠand growth traitsThe results showed that two kinds of genotypes CC and CD exist in GH gene ExonⅠ, sequencing results showed that CD and CC has a mutation C→T in 372th base, the C allele was dominated in this loci, and the group was in Hardy-Weinberg extreme imbalance(P<0.001). Two genotype of GH gene exonⅠhas significant difference on two-month-old body length (P<0.05), CC>CD; on birth weight and other growth traits were no significant differences (P>0.05).3 Associations of polymorphisms of Haimen goat GH gene exonⅡand growth traitsGH gene ExonⅡalso has two genetypes EE and EF, EFand EE has a mutation G→A in 782th base, E allele was dominated in this loci, and the group was in Hardy-Weinberg extreme imbalance(P<0.001). GH gene exonⅡof the two genotypes on two months weight and two months withers height were extremely significant differences (P<0.01), EE>EF; It was significantly different between the genetypes and 2-month-old body length and Cannon circumference (P<0.05), EE>EF; for other growth traits were no significant differences (P>0.05).4 Associations of polymorphisms of Haimen goat BMP15 gene IntronⅡand litter sizeThe results showed that the BMP15 gene IntronⅡwas found only one genetype HH, nopolymorphisms, has no significant on litter size of Haimen goats. It indicated that this site at Haimen goat groups have reached a homozygous, or the mutation site does not exist.
