Sorbifolia belongs to Sapindaceae and sorbifolia case, a deciduous shrub or small tree, it is an unique woody medicinal and oil tree in China. Sorbifolia is widely distributed in the northern of China. Sorbifolia has strong ability to endure drought and cold. Sorbifolia leaves, stems, flowers and husk contain a variety of active ingredients. The use of plant tissue technology can solve the difficult process of sorbifolia breeding. Meanwhile, genetic engineering research is based on tissue culture.This study used sorbifolia embryo as explants. Based on the previous studies and draw on its same family plants as well as other common oilseeds tissue culture techniques, sorbifolia tissue culture regeneration system was established. The main results are as follows:1. During the callus induction process, I compared the impact of MS,1/2MS, B5and WPM, the several basic medium, on callus induction, then found that MS is the most suitable basic medium for callus induction. Contrasting the effects of2,4-D,6-BA and NAA on callus induction, the most suitable medium and hormone combination for sorbifolia embryo callus induction is MS+2,4-D1.Omg/L, induction rate reached96.7%.2. In the callus subculture process, different concentration of6-BA have been used. The result is that the optimal medium for callus subculture is MS+2,4-D 0.5mg/L+NAA0.2mg/L+6-BA0.2mg/L3. The best bud induction medium for sorbifolia is MS+6-BA2.0mg/L+NAA0.5mg/L, the induction rate is64.4%.6-BAhas stronger ability to induce adventitious buds than KT.4.1/2MS medium is more suitable than WPM for sorbifolia rooting, IB A is more effective than other hormones in rooting promotion. Adventitious buds in1/2MS+IBA1.Omg/L+NAA0.5mg/L, the rooting rate is55.6%.
