Object: Lily is the dry scale leaf of Lilium lancifolium Thunb.,L. brownii F.E.Brown var. viridulum Baker and L. pumilum DC. originated from Liliaceae Lilium, which is not only a common Chinese medicine but also a staple trueborn medicine in Hunan. Many methods ,such as morphological, physical and chemical identification, have been applied in the identification on Lily. There only one relevance studies report has been reported in identification of Chinese medicine Lily on molecular evidence yet. The technique of random amplified polymorphic DNA(RAPD) ,as a DNA molecular marker , was used in this study , in which included not only Lily from different habitats in Hunan, Guangxi, Jiangsu, Gansu and Zhejiang but also two different generic plant: Lilium and Cardiocrinum. We have provided the sufficient experiment basis to the new method of identifying different lilies and discussed the applicaton of the technology of molecular biology on identifying the species or cultivars and analyzing the genetic relationship among them. Methods: Adopting the RAPD technology to analyze the lily scale leaf's genic DNA ; using the gel imaging analysis system to establish DNA fingerprint; using the UPGMA in the NTSYS programs to analyze the genic relationship of every sample.The results and conclusions : (1) The RAPD reaction system suitable for Lily was established; (2) 35 primers were screened from 120 random 10-mer primers, and 769 DNA bands were amplified ,767 of which (99.7%) were polymorphics; (3)The DNA fingerprints via analyzing the electrophoresis image were obtained by using the gel imaging analysis system; (4) The RAPD results showed the same conclusions as those obtained from the studies on morphology and cytology; (5) The RAPD technology provided the biological essential of trueborn medicine in the molecular level.It was the first time to analyze the genetic relationship and identify different lilies with the RAPD technology and to build the DNA fingerprint, which is a beneficial try to and a new method in Chinese traditional medicine identification.
