Melilotoides ruthenica is a most important forage species. This paper taking 3 new strains of Melilotoides ruthenica (L.) Sojak (90-36(A), 93-21(B) and 00-61(C))as materials, which were screened by Grassland Research Institute of Chinese Academy of Agricultural Science. Firstly, using the methods of tissue culture, include normal callus induction, normal callus differentiation and yong stem culture studied the quickly extend techniques of Melilotoides ruthenica strains. These techniques is of service to their factory seedling culture. At the same time, seed yield components and quantity characteristics of new strains were tested and analysed. Third, useing strain 00-61(C) as material, studied the influence of ordinary manure, N, P, K, and tiny manure Mo, B, Cu, Zn on the seed yield components and the seed yield, explore the fertilize effect of new strains. The results were as follow:1)The feasible medium of callus inducing of cotyledon of three Melilotoides ruthenica strains is MS+2,4-D1.0 mg/L+KT0.2mg/L,the feasible medium of callus inducing medium of hypocotyls of strain A and C is SH+2,4-D 0.5mg/L + 6-BA0.7mg/L,the feasible medium of callus inducing of hypocotyls of strain B is SH+2,4-D0.5mg/L+6-BA0.5mg/L;The feastible medium of normal callus differentia- tion of hypocotyls of all three strains is MS+KT1mg/L+6-BA1mg/L;The rooting medium of all three strains is MS medium; The subculture, proliferation and rooting medium of yong sterm of all three strains is SH+NAA0.1mg/L. The propagation coefficient through callus induction of strain A,B and C are 8.47,7.83 and 5.65, respectively. The propagation coefficient through yong sterms of strain A,B and C are 6.91, 6.52 and 5.82, respectively.2) 0.5mg/L 2,4-D plus 0.5~0.7 mg/L 6-BA is the good concentration for the hypocotyls callus inducing, at this concentration on the SH medium, the average normal callus induction rate of three strains is 92.7%. While,1mg/L KT plus 1mg/L 6-BA is the good concentration for the callus differentiation, the callus differentiation rate is 31.51% on this concentration. The difference of callus induction rate and callus differentiation rate of three strains are not significant.3) The effective shoots/m2 of strains A, B and C are 269, 247, 530; The inflorescence/ effective shoots are 68.69, 48.96, 23.79; The flowers/ inflorescence are 5.79, 9.91, 8.79; The pods/ inflorescence are 2.66, 6.24, 3.78; The ovules/pod are 4.05, 3.61, 4.23; The seeds/pod are 2.09, 3.45, 3.15; The 1000-seed weight (TKW) are 3.19g, 1.87g, 2.54g, respectively. The most seed yield affecting components shown a formal distribution pattern.4) The correlation coefficients of actual seed yield with expression seed yield, potential seed yield and TKW at 0.01 level were 0.900**, 0.917** and 0.950**; The correlation coefficients of actual seed yield with effective shoots/m2, inflorescence/ effective shoots and seeds/pod were 0.033, 0.594 and 0.301. The actual seed yield with seed yield affecting components regression equation was Y=124.035+14.318X7+0.014X2 (X2: inflorescences/ effective shoot, X7: TKW ).Melilotoides ruthenica. has a hight flower dropping rate, the pod dropping rate is lower than the flower dropping rate , but it has a little bud dropping rate .5)All treatments of fertilizer can produce significance effect to potential seed yield, expression seed yield and actual seed yield on strain C of M. ruthenica. The treatment of 90 kg/hm2 N,220 kg/hm2 P2O5,60 kg/hm2 K2O is better than other treatments, which increased 25.87% of actual seed yield. N, P and K have significant positive effect on effective shoots/m2, inflorescences/effective shoots, flowers/ inflorescence, pods/ inflorescence, ovules/pod, seeds/pod and TKW.6)Mo has high effection to improving the seed production and the seed yield effecting components, second was Zn, which as micro-nutrients in treated. They increased seed production by enhanced the seed yield effecting components.
