Study On Agronomic Trait And Genetic Analysis Of Hybrid And Their Progenies Detected By SRAP Markers In Annual Ryegrass

Abstract/Summary:
Eight Lolium multiflorum germplasm materials were firstly tested Southwest China. The 70 samples containing 8 annual ryegrass hybrid combinations and their 62 progenies were analyzed using sequence-related amplified polymorphism (SRAP) molecular markers to study the polymorphism and detect true and fake hybrid seeds. As ten kinds of annual ryegrass were used as control, we compared and evaluated the phenophase, forage yield, seed yield and the ratio of stem and leaf of "YA97-17" which is the new line of annual ryegrass in Sichuan, Ya'an, and Chongqing Rongchang, and analyzed its adaptability in Southwest China. It provides the theory foundation for the deep research of the breeding and the reasonal exploitation and use of the Lolium multiflorum germplasm. The main results are as follows:1) Ten primer pairs produced 101 polymorphic bands, with an average of 10.1 bands per primer pair. The percentage of polymorphic bands averaged 77.1%. The results indicated that the samples have relatively higher molecular genetic polymorphism. So it is feasible to identify whether it is true hybrid. 62 hybrids generated from 12 different hybrid combinations were identified by 9 SRAP primer pairs . The results showed that: 48 progenies having the male parents' bands were true hybrid and other 14 progenies having no male parents bands were false hybrid. At the same time,the bands of hybrid are not the sum of the parents' bands, there were new bands presented and some bands are missing. And the bands of the same hybrid combination are different even their primer pairs of SRAP are the same. To achieve more exact and general results, the research identified the same hybrid of one hybrid combination using different SRAP primer pairs. All the research showed hybrid breeding of Lolium multiflorum resulted in the recombination of gene and abundant variations. And all these provided foundation for hybrid seed cultivation and selecting. The hybrid identification laid a foundation for further selecting and breeding hybrid .2) It indicates that "YA97-17" appeared to have strong acclimatization through one year region trial. The quality and palatability of "YA97-17" are excellent. From the testing results of the 11 breed of Lolium multiflorum , it can be tested for 3 times. And "Chenqu" has the most amount of dry forage yield in Ya'an, and achieved 34736.80kg/hm~2, and the second was "Jianbao" , and the third was the new line "YA97-17" , achieved 34158.96 kg/hm~2; The least amount was "Gulf. "Chenqu" had the most amount of fresh forage yield, achieved 176328.70kg/hm~2, the second was "Jianbao" , the third was the new "YA97-17" achieved 171883.82kg/hm~2. The least amount was "Gulf". In Rongchang, the new "YA97-17" has the most amount of dry forage yield, and achieved 160106.26kg/bm~2; the second was "Barspectra"; the third was "Abode", achieved 171883.82kg/hm~2; The least amount was "Liaoyuan". The average yield of fresh grasses of the two regions more than "Changjiang -2" and "Tetrtagord" were increasing over 8.1 % , 8.99 %, the average yield of dry grasses of two regions more than "Changjiang -2" and "Tetrtagord" were increasing over 8.22 % and 8.89 % , so the yeild had obvious advantage. In conclusion, "YA97-17" grows better in southern subtropic of China, and is an excellent new line of Lolium multiflorum.
