Swine streptococcosus disease is caused by Swine Streptococcus infections.It is zoonosis and distributed in a wide range in the world,which is the second-class animal disease.Because of it's excessive serotypes and complex antigen structures,the clinical characteristics are different. For example,septicaemia, meningitis, endocarditis ,pneumonia, , polyserositisand arthritis. It's very difficult to diagnose.It has been reported that this kind of disease occurred in more than 20 states, including China. So,finding a simple and fast way to choose certain typical strains by using them producing vaccine is in need.This study is to develop the genotyping method of randomly amplified polymorphic DNA(RAPD) for it, Screening optimization RAPD conditions.Statistics from various strains of the assault's genetic distance,and cluster analysis,drawing relationship tree.The results show that: A total of 791 pig nasal fluid and submandibular lymph nodes, collected From slaughterhouses in Luoyang, Henan and Lanzhou, Gansu . 13 strain s of pathogenic streptococcus of swine isolated from samples. By bacterial isolation and culture, biochemical tests, Lancefild grouping A-G diagnostic reagents identification, Tests the results were 5,7 and 1 strains for group C,D and F respectively. Drug sensitive test results showed that the isolates to ampicillin, vancomycin, chloramphenicol, and Ceftriaxone Sodium highly sensitive;to gentamicin, amikacin, erythromycin, ofloxacin, ciprofloxacin hydrochloridel moderately susceptible; have already developed resistance to penicillin, compound sulfamethoxazole, streptomycin, norfloxacin, Clindamycin, norfloxacin, tetracycline.Random amplified polymorphic DNA(RAPD)is used for analysising the isolated strains, to establish the best RAPD response system.In this experiment 5 prinmers were selected among 20 ones. This method can amplify genomic DNA segment by random primers, 13 Streptococcus suis will be divided into eight groups clustering. The results of the present study have showed a relative genetic diversity among the different isolates of streptococcus. Serum of different groups with different strains of fingerprints, The same fingerprint of the serum group there are also differences. RAPD is better than the traditional method to reflect the relationship between pathogen strains.RAPD is a simple and fast techmique, so it has been applied to typing of Streptococcus since it was invented. It provides a theoretical basis for the Swine strep tococcosis control and epidemiological investigation at the molecular level.
