In this paper, 13 tea plant cultivars were evaluated using RAPD markers. The result showed:1. PCR optimum amplification of genomic DNA as follows: 94℃ for 40 s; 45 cycles of 94 for 30 s, 37℃ for 60 s, 72℃ for 1 min; 72℃ for 10 min. The PCR reaction was performed in a final of 25ul containing 1 X Tag polymerase buffer, 1 unit of Tag polymerase, 200uM of each dNTPs, 0.2uM of random primer, 2.5mM MgCl2 and 50ng of total genomic DNA.2. RAPD-PCR using 20 random primers producted 125 readable and reproducible binary patterns (62 constant, 63 variable), and the size of amplification products ranged from 0.3kb to 3.0kb, which were used to calculate the genetic diatances and to construct the dendrogram. Out of the 125 bands, 63 bands(about 50.4%) were found to be polymorhphic. It suggested the diversity detected among the tea cultivars was relatively low.3. The results of cluster analysis revealed that three major groups could be recognized. Group 1 included "Moxiang", "Benshan", "Ruanzhiwulong" and "Ivyaguanyin". Group 2 comprised of "Hongyafushou", "Dayewulong"; Group 3 included "Yuanyeguanyin", "Changyeguanyin". However, "Xingren", "Tieguanyin 1", "Ivyafushou", "Hongguwulong" and "Tieguanyin 2" were different from above cultivars. The genetic similarity varied from 0.74 to 0.91, with the average of 0.8516. The genetic distance values were between 0.3011 and 0.0943, with the average of 0.1607.The comparison of Tieguanyin 1 with the other 12 cultivars showed that the genetic distance between Ivyafushou and Tieguanyin 1 was the longest and the largest, However, the genetic distance between Benshan and Tieguanyin 1 was the smallest.
