The polymorphisms of growth hormone (GH) gene in three yak populations were analyzed by PCR-SSCP. The five exons and parts of introns of growth hormone (GH) gene were detected in the three groups. It aims to get molecular genetics information and find the growth character correlation relevance to the molecular. The objectives of the present study can provide the scientific basis for the protection development and using of the special heredity resources-yak. The results as following:The products amplified by exon1,exon2 and exon4 display polymorphism in the three groups. The result of PCR-SSCP polymorphism examination of the GH-P3:It was controlled by alleles gene of A and B. There were three genotypes,namely AA,BB,AB in intron 3. Cloning and sequencing indicated that there was one single nucleotide mutation (T/C) at 1 694bp in intron3.Frequency of A allele was 0.8342,0.9028 and 0.8958. The result of PCR-SSCP polymorphism examination of the GH-P5:It was controlled by alleles gene of C and D. There was one single nucleotide (A/G)at 2154bp, and this mutation resulted in an amino acid change:glycine/serine of the Gannan yak population. One single nucleotide mutation(G /T) at 2373bp of the Tianzhu white Yak population,and it did not induce the changes of encoding amino acid. There was no polymorphism in the Qinghai yak. Statistical results showed that the GH SSCP sites in 3 populations of the yak were at Hardy-Weinberg disequilibrium (P<0.05),PIC was low polymorphism.
