Hezuo pig is a small, slow-growing breeds, grazing strong performance, withfatigue, fear exposure,and snow, and other special properties. The polymorphism, allelenumber, nucleotide polymorphism sites, amino acid polymorphism sites, the genetic relationship andevolutionary significance of the alleles of the SLA-DQA gene exon2and4in Hezuo pigs were studied.Selection of PCR-SSCP,439of Hezuo pig SLA-DQA gene polymorphism in exon2and4were detected; each allele sequencing groups generated internal variability and analyzesequence data, while building homologous tree in order to further clarify the Hezuo pigbetween SLA-DQA gene allele and the plains breeds, the genetic relationship of foreignbreeds. Exon2was found in seven new alleles in Hezuo pig SLA-DQA, the total of18nucleotide polymorphic sites,10amino acids of polymorphic loci; Hezuo pig SLA-DQAfound in exon4the four new alleles of six nucleotide polymorphism loci, four aminoacids of polymorphic loci, indicating that Hezuo pig SLA-DQA exon2and4, with awealth of polymorphism within populations may bears richer genetic resources.Constructed by SLA-DQA gene homology tree showed Hezuo pig SLA-DQA exon2and4may initially by a mutation in one allele differentiation into a large class of genes;Hezuo pig SLA-DQA exon2and4common sequence with the various breeds ofSLA-DQA exon2and4sequences with high homology, indicating that these breeds ofSLA-DQA exon2and4gene may be derived from their differences before the earliest ofancestors of the original sequence; newly discovered seven SLA-DQA exon2alleles,may be provided by closer genetic relationship between two mutant alleles produce.
