| Peste des petits ruminants (PPR), casued by peste des petits ruminants virus, is an acute feverish and highly contagious disease. it is the hazardousextremely serious disease, and classified as group A disease by Office International des Epizooties(OIE)and group first disease in China. In 2007, the PPR epidemic occurs in our country , the continually recurring of PPR in our surrounding countries and domestic puts forward a new demand for the prevention and control animal epidemic of our country . The diagnosis and prevention of PPRS becomes important research field.In this study, the eukaryotic expression vectors were constructed containing H gene and IFN-γas adjuvant cytokine .and used to research the expression of the target gene in the BHK-21 cells and the ability of inducing humoral and cellular immunity.the results is as follows:1 .The clone of PPRV H gene, construction of eukaryotic expression vectors containing H gene and expressed in BHK-21 cells. the target gene were digested by Not I and Xhol and inserted into pBude4.1 and pBud-IFN The expression vector pBud-H and pBud-IFN-H were transfected into the BHK-21 cells by liposome mediated method.. , immunofluorescent test showed transcription and expression of the target genes in the BHK-21 cells.3 The research of DNA vaccine if it can inducing the Humoral and cellular immunity of mouse. pBud-H and pBud-IFN respectively, while costimulatory molecule IFN-γas immunoadjuvant gene. 4-week-old female healthy BALB/c mice were respectively immunized, and a booster vaccination was given two weeks later. Kinetic tests were carried out by ELISA and MTT assays. Results show that humoral and cellular immunoresponse were not only elicited by pBud-H and pBud-IFN-H DNA vaccines, but also humoral immunoresponse elicited by pBud-IFN-H was stronger than that by pBud-H.Briefly, DNA vaccines can induce stronger cellular immunoresponse and weaker humoral immunoresponse than what PPRV purified antigen immune. |
