170 blood samples(Gansu red deer,Qinghai red deer,TianQing F1 red deer, TianQing F2 red deer and TarimGan F1 red deer) were amplified and detected,using microsatellite DNA markers.Analyzed alleles size and frequency of each gene loci; conducted genetic heterozygosis(He),polymorphism information content(PIC), Nei's genetic distance(D_A) and Nei's Standard Genetic Distance(D_S);dendrogram was constructed using the neighbor-joining and UPGMA methods;analyzed 8 locus gene frequency principal constituent for the 5 red deer populations.Studied genetic diversity of Gansu red deer and genetic relationship between Qinghal red deer.The results showed that:10 microsatellites loci were choosed to amplify 170 blood samples,8 of them(BM203,BM888,BMS1248,TGLA226,BM1818,BM3628, BMC1009 and BM1225) were amplified successfully;2 of them(BM2320 and MAF70) were amplified unsuccessfully.Using 8 microsatellites loci to detect genome DNA of 170 blood samples,got 72 alleles,the average number is 9 in each loci. BM888 and BMC1009 have the highest number(12);TGLA226 has the lowest number(5).The average allele frequency of the 8 microsatellites loci distribution is similar between Gansu red deer and Qinghai red deer,7 of them have the same preponderant alleles.The 8 microsatellites loci showed high PIC value and polymorphism of Gansu red deer,the mean expectation heterozygosity was 0.7948; the mean content of polymorphism information was 0.7566.Phylogenetic analysis showed that Nei's genetic distance(D_A) values and Nei's standard genetic distance(D_S) values between Gansu red deer and Qinghai red deer both were smaller,i.e.0.0464 and 0.0227,respectively.Phylogenetictree illustrated the same conclusion.Principal component analyses(PCA) indicated that Gansu red deer and Qinghai red deer populations are separated from the other 3 red deer populations.The Gansu red deer population showed high expectation heterozygosity and polymorphism information, showing rich genetic diversity.The 8 microsatellites loci could be used as genetic diversity analysis of red deer.Genetic Differentiation based on molecular level between Gansu red deer and Qinghai red deer are inconspicuousness.In this study, the results showed that,Gansu red deer and Qinghai red deer populations show similar relationship,using microsatellite analysis.
