| Chrysanthemum morifolium Bozhou is one of very high valued medicinal chrysanthemums, and its flowers contain flavonoids, such as adenine and composition, there sparse wind radiation, clear liver clear vison, the role of solution is a combination of medicinal and beverage in medicine,and also the main export products. In recent years, with the efficacy of medicinal Chrysanthemum continuous research and development, the market demand increasing for its number,species and the quality, but traditional breeding methods and natural variability on the way to breeding new varieties of low propagation coefficient, the efficiency of industrial production is not high, has been unable to meet the needs of production, in particular the sub-yield after many generations, often causing the degradation of caste, and even virus accumulation, yield and quality declined significantly. Therefore the tissue culture virus-free technology for Chrysanthemum morifolium Bozhou is a key process to solve this problem.This experiment takes tip as explant, the choice of explants, multiple shoot clumps induction, the proliferation of multiple shoot clumps, take root, and the process of transplanting various influencing factorsasepsis, and heat treatment and untreatment transplanting seedlings' virus-free and physiological index were studied. the main contents focus on the following aspects, such as basic mediums and raising modes, the combination and density of hormone, different from the choice of virus-free technology and physiological index. The Orthogonal Design is adopted to arrange experiment. The results showed as following:1.Taking Chrysanthemum morifolium Bozhou tip as explant, the best medium for: MS+6-BA3.0mgL-1 + NAA0.2mgL-1 + sucrose 40gL-1, the combination of germination rate of 75.0 percent, an average of differentiation rate of 1.40.When non-leaf primordium tip, survival rate is very low only 20 percent; when the shoot tips with leaf primordium size is 1.0mm, the tip can be as high as 66.7 percent survival rate.2.The main factors of multiplication of bud clumps are 6-BA,NAA and sucrose. The results showed that: Chrysanthemum morifolium Bozhou subculture optimal combination of the proliferation of cultured is A1B1C1, the most appropriate medium is MS+6-BA0.5mgL-1 + NAA0.1mgL-1 + sucrose20gL-1, the best bud proliferation.3. Within the scope of the trial, in the medium MS + NAA0.5mgL-1+ sucrose 30gL-1 the rooting results are the best.4. When transplanting, the first to go through 5-6days of Lianmiao period, then shade in the indoor ventilation, the average humidity of 70 percent to 80 percent of the greenhouse transplanting, sand and perlite survival rate was 100 percent, and the best seedling growth.5. Measured indicators part of heat treatment, non-heat treatment and the untreated Chrysanthemum morifolium Bozhou. The results showed that: The total flavonoids, chlorogenic acid and chlorophyll content of the three indicators are more obvious physiological differences, the second to deal with the Chrysanthemum morifolium Bozhou flowers share of all three indicators were higher than those of untreated Chrysanthemum morifolium Bozhou, that may suggest the different resistance to plant disease .6. The agronomic characters and the flowering chrysanthemum flowers of heat treatment, non-heat treatment and untreated Chrysanthemum morifolium Bozhou had been determined and evaluated. The results revealed that comparing with those of untreated Chrysanthemum morifolium Bozhou plants,the agronomic characters of the second to deal with the Chrysanthemum morifolium Bozhou plants had changed significantly,and the leaves larger.The diameter of capitulum and the numbers of ligulate petals were increased noticeably.In a word,exterior characters of capitulum in the second to deal with the Chrysanthemum morifolium Bozhou plants were superior to untreated plants.7. Direct observation and the instructions were detected virus on Chrysanthemum morifolium Bozhou of the second heat treatment plant and the second non-heat-treatment was detected virus. The results showed that: the second heat-treatment samples with a TAV, and three with PVY; and the second non-treatment samples with three TAV and 4 with PVY. |
