As a considerable component during In Vitro Culture (IVC) of oocyte, sulfhydryl were used some one, but there were no system studies in effect of kinds and additive dose of sulfhydryl on In Vitro Maturation(IVM) of oocyte. The purpose of this experiment was to study the effect of cysteine, GSH,β-ME and cysteamine on the IVM of Yanbian Yellow Cattle oocyte and development of somatic cell clone embryo, so as to increase the clone efficiency of Yanbian Yellow Cattle somatic cell. Recover the oocytes by sucking in slaughtered ovaries, then carry out IVM in sulfhydryl of different kind and concentration, finally carried out IVC with the reconstructed embryos which were constituted by nuclear transfer of mature oocytes. The results showed as follow:In the study of effect of cysteine on IVM of Yanbian Yellow Cattle oocyte with different concentration in IVM fluid,0.56 mmol/L group was significantly higher than other groups (P<0.05), so it is the most suitable concentration of cysteine added alone for Yanbian Yellow Cattle IVM of oocyte.In the study of effect of GSH on IVM of Yanbian Yellow Cattle oocyte with different concentration in IVM fluid,1.0 mmol/L group and 1.5 mmol /L group had no significant difference (P>0.05), but 1.0 mmol/L group was significantly higher than other groups (P<0.05),1.0 mmol/L group is the highest. so it is the most suitable concentration of GSH added alone for Yanbian Yellow Cattle IVM of oocyte.In the study of effect ofβ-ME on IVM of Yanbian Yellow Cattle oocyte with different concentration in IVM fluid, there was no significant difference between 100 umol/L group and 150 umol/L group (P>0.05), but 100 umol/L group significantly higher than other groups (P<0.05), so it is the most suitable concentration ofβ-ME added alone for Yanbian Yellow Cattle IVM of oocyte.In the study of effect of cysteamine on IVM of Yanbian Yellow Cattle oocyte with different concentration in IVM fluid,100 umol/L group and 50 umol/L group,150 umol/L group had no significant difference(P>0.05), but 100 umol/L group significantly higher than control group and 200umol/L group (P<0.05), so it is the most suitable concentration of cysteamine added alone for Yanbian Yellow Cattle IVM of oocyte.In the study of effect of adding the determined-concentration medicine on IVM of Yanbian Yellow Cattle oocyte in IVM fluid, cysteine combinated cysteamine group and cysteine combinatedβ-ME group had no significant difference(P>0.05), but both groups significantly higher than control group and GSH group (P<0.05).so determined quantity of cysteine combinated cysteamine was the most suitable for Yanbian Yellow Cattle IVM of oocyte.In the study of effect of cysteine on efficiency of Yanbian Yellow Cattle somatic cell clone with different concentration in IVM fluid, the 0.56 mmol/L group had no significant difference, contrasted with 0.28 mmol/L group,0.84 mmol/L group (P>0.05), but significantly higher than control group and 1.12 mmol/L group (P<0.05) in rate of cleavage, so 0.56 mmol/L group was the highest; in rate of blastula development, there was no significant differences among groups added medicine (P>0.05), but 0.56 mmol/L group significantly higher than control group (P<0.05). so it is the most suitable concentration of cysteine added alone for Yanbian Yellow Cattle somatic cell clone.In the study of effect of GSH on efficiency of Yanbian Yellow Cattle somatic cell clone with different concentration in IVM fluid. In rate of cleavage, there was no significant difference between 1.0 mmol/L group and 1.5 mmol/L group t (P>0.05), but significantly higher than other groups (P<0.05), so 1.0 mmol/L group is the highest; in rate of blastula development,groups added medicine had no significant difference (P>0.05), but 1.0 mmol/L group significantly higher than control group (P<0.05).so it is the most suitable concentration of GSH added alone for Yanbian Yellow Cattle somatic cell clone.In the study of effect ofβ-ME on efficiency of Yanbian Yellow Cattle somatic cell clone with different concentration in IVM fluid.
