The acetylation of histone takes part in mediating various kinds of chromatin that in the process of mitosisi. In the nomal procrdure of fecundation and embryo development, in which the epigenetic,including the histone acetylation, throughout their lives.Some research improved that the epigenetic of donor nucleus in the receptor oocytes is imcompleted and abnormous. The object of this research is to explore the relationship between the hitone acetylaion and cloning embryos,and also for make further improvement on expounding the mechanization about the development of cloning embryos,thus to improve the efficiency of clone. This experiment made a thorough inquiry on the effect of histone deacetylase inhibitor (TSA) on the cloning reorganization embryos of Yanbian cattles.This research adopted the histone deacetylase inhibitor ttichostatin A to examine the influence of TSA in different acting time and different concentration,the result shows as follow.1. In the study of effect of TSA on IVM of Yanbian Yellow Cattle A-grade oocyte and efficiency of Yanbian Yellow Cattle somatic cell clone with different concentration in IVM fluid,50 nmol/L group is not significantly higher than the control group-in rate of oocyte maturation,but significantly higher than other groups(P<0.05) in rate of blastula development, so it is-the suitable concentration of TSA for Yanbian Yellow Cattle IVM of oocyte and omatic cell clone as addition.2. In the study of effect of TSA on IVM of Yanbian Yellow Cattle B-grade oocyte and efficiency of Yanbian Yellow Cattle somatic cell clone with different concentration in IVM fluid,50nmol/L group was better than other groups in every index with significant difference(P<0.05), so it is the suitable concentration of TSA for Yanbian Yellow Cattle IVM of oocyte and omatic cell3. Motieeastaid(?)neffect of TSA on IVM of Yanbian Yellow Cattle oocyte and efficiency of Yanbian Yellow Cattle somatic cell clone with different acting time in IVM fluid,22h group was better than other groups in embryo development index with significant difference(P<0.05), so it is the suitable acting time of TSA for Yanbian Yellow Cattle IVM of oocyte.4. In the study of effect of TSA on efficiency of Yanbian Yellow Cattle somatic cell clone with different concentration in activation medium,50nmol/L group was better than other groups in the index of cleavage and blastula with significant difference(P>0.05), so it is the suitable concentration of TSA for Yanbian Yellow Cattle activation stage of somatic cell clone as addition alone.5. In the study of effect of TSA on efficiency of Yanbian Cattle somatic cell clone with different concentration in after-nuclear transform IVC medium,the result shows that the rate of cleavage and blastula is neither higher than any other groups(P>0.05),so the addition of TSA in this stage has no positive effect.6. In the study of effect of TSA on efficiency of Yanbian Yellow Cattle somatic cell clone with different concentration in IVC medium,50nmol/L group both better than other groups with significant difference in rate of cleavage and blastula development(P<0.05), so it is the suitable concentration of TSA for Yanbian Yellow Cattle of somatic cell clone as addition.7. In the study of effect of TSA on efficiency of Yanbian Yellow Cattle somatic cell clone with different acting time in IVC medium,24h group was better than other groups in embryo development index with significant difference(P<0.05), so it is the suitable acting time of TSA for Yanbian Yellow Cattle IVM of oocyte...
