| To know the degree of genetic difference in DNA lever among 36 potato varieties, the polymorphism among the tested plants were analyzed based on SSR molecular marker. The results showed that:1. The optimal concentrations of four main Factors in the potato SSR-PCR reaction system were selected: 2.0mmol/L Mg2+, 2.0mmol/L dNTP, 0.2U Taq and 2.0μmol/L primer.2.The10 pairs suitable primers were selected: STM11,06,STACCA3,SSR111,S180, S7,POTM1-2,STINHWI,STIIKA,STU6SNRN,S192.3. 301 sites of SSR markers were amplified in the genomic DNA of 36 potato varieties with 10 pairs suitable primers, and the percentage of polymorphism sites reached 71.1%. The SSR fingerprinting of each primer distinguish 36 potato varieties clearly, so it could be the molecular evidence to identify the potato varieties.4. The genetic distance(GD) of 36 potato varieties were ranged from 0.3333 to 0.8500. Using the GD value of 0.57 as threshold, tested plants could be clustered into 7 groups. Analysis of genetic similarity coefficient (GS) showed that: the GS values of most potato varieties were above 0.6, so their genetic base was relatively narrow. |
