The objectives of this study are to establish the parentage testing system of Chinese Simmental and analyze the parentage in study population.1626 Chinese Simmental, including 1311 cows and 315 sires are genotyped for 12 microsatellite markers, and evaluating the genetic information and exclusion probability of these markers. Exclusion-based and likelihood-based methods are used to infer parentage in the study population of Chinese Simmental. The main results are as follows:1. The average of heterozygosity , polymorphism content information and alleles number are 0.735,0.705 and 12.42, respectively. The difference between expected heterozygosity and observed heterozygosity ranged from 0.003 to 0.097 confirming a high polymorphism of each locus. Their combined exclusion probability reached 0.9979, 0.9999 and 0.99999 in three different categories, which made them suitable and sufficiently accurate for parentage testing.2. The system of 12 microsatellite markers for parentage testing is established. They are: ETH10, BM2113, BM1824, ETH225, TGLA126, TGLA122, INRA032, SPS115, INRA037, CSSM66, TGLA53 and HAUT27.3. Analyzed parentage of 1626 Chinese Simmental with the parentage testing system in the study population. Combined with exclusion-based method and Likelihood-based method to assign parent of the offspring . As a result, 406 offspring found their father,386 found their mother and 80 found their parents with confidence above 80%. These results providing the basic information for animal breeding program.
