The whole group of Liangshan Semi-fine wool sheep of nuclear breeding farm were used in this experiment. Therty six microsatellite DNA markers contained in three multiplex systems which distributed in four different chromosomes,the first group contained 17 markers,the second one contained 10 markers and the third one contained 9 markers. Allele frequencies were estimated and the x2 was so computed ,values of polymorphic information content and heterozygosity and the probability of exclusion of every loci and the combined probability of exclusion were calculated based on allele frequencies. Experimental pedigree of a sire labeled H1G4003 was fouded relied on the 36 loci,and 7 half-sib pedigree were also founded in the same time.Every loci analyzed was polymorphic and the number of average allele number was 8.8,distribution of allele frequencies of every loci conformed to the Hardy-Weinberg theory. The average heterozygosities of three sets of microsatellite marker were 0.66297 and 0.72615 and 0.74166,their probability of exclusion were 0.99999999 and 0.9999999 and 0.999999 respectively,the probability of exclusion of 9 markers was slightly higher than published. We foud that pedigree was correct except few of cases through comparison between experimental pedigree and pedigree,and the records of pedigree of 1998 were all correct,they can used in parentage testing. Number of seven half-sib pedigrees were founded through the method of parentage exclusion conform to the analysis requirment of QTL.
