Amino acids are an important component in vitro culture medium, the current application of the culture medium contains a variety of essential/non-essential acid, but few have combined for a single amino acid or several amino acids on embryo development to add the impact of research reports. The present work studied the culture medium to add L-glutamine, taurine, glutamic acid, alanine, proline, glycine, arginine and glutamine and taurine added to the restructuring of the joint cleavage,8-16C embryo development, blastocyst development impact, with a view from which to explore the possibility of amino acids to improve the efficiency of somatic cell cloning Yanbian cattle. The experiment was in vitro culture medium were added over several different types of different concentrations of amino acids, on the reorganization of somatic cell cloning embryos of Yanbian cattle cultured in vitro, and then observe the reconstructed embryos had developed.Experimental results are as follows:1. In vitro culture medium added with different concentrations of glutamine on the Yanbian cattle embryo development in somatic cell cloning re-test the effects of adding 1.0 mmol/L glutamine group cleavage rate of reconstructed embryos,8-16C growth rates are higher than other concentrations, blastocyst rate was significantly higher than other concentrations (P<0.05), so set 1.0 mmol/L L-glutamine was alone when the most suitable for Yanbian cattle oocytes somatic cell cloned embryo development re-The optimum concentration.2. In vitro culture medium added with different concentrations of taurine on the Yanbian cattle embryo development in somatic cell cloning re-test the effects, add 7mmol/L and 14 mmol/L taurine group was higher than that re-cleavage embryos Other concentrations,8-16C growth rate and blastocyst rate was significantly higher than in other concentrations (P<0.05), but 7mmol/L group in the cleavage rate of reconstructed embryos,8-16C growth rate, blastocyst rate on were higher than 14mmol/L group. Therefore, to determine 7mmol/L is the most suitable when taurine alone Yanbian cattle oocytes somatic cell cloned embryo development in reorganization of the optimal concentration.3. In vitro culture medium with different concentrations of L-arginine added to the Yanbian cattle embryo development in somatic cell cloning re-test the impact of various concentrations between the cleavage rate of reconstructed embryos and 8-16C no significant difference in growth rate, blastocyst development on the rate of 0.61mmol/L concentrations were significantly higher (P<0.05), all experimental groups no significant difference between, but 0.61mmol/ L group blastocyst rates higher than other experimental group, so determining 0.61mmol/L, L-arginine alone, when the most suitable Yanbian cattle oocytes somatic cell cloned embryo development in reorganization of the optimal concentration.6. In vitro culture medium with different concentrations of glycine added to the Yanbian cattle embryo development in somatic cell cloning re-test the impact of various concentrations between the cleavage rate of reconstructed embryos and 8-16C no significant difference in growth rate, blastocyst development on the rate of 7mmol/L group were significantly higher than that and the highest concentrations (21mmol/L. In vitro culture medium with different concentrations of proline added to the Yanbian cattle embryo development in somatic cell cloning re-test the impact of each group of the cleavage rate,8-16C growth rate, blastocyst rate was no significant difference. but the 0.143mmol/L concentration group than other groups more suitable for somatic cell cloned cattle in Yanbian reconstructed embryos in vitro.
