[Purpose] The foundation of this study was the injury to the cell generated by ROS. A variety of antioxidants in different concentration were added in culture media to evaluate their effects on embryo developmental potentiality, and decide the optimal density for Somatic Cell Nuclear Transfer embryo in vitro culture in YanBian Yellow Cattle. Thereby the purpose of this article was to solve the problem of development block caused by existence of peroxides, decrease oxidative stress and enhance the efficiency of nuclear transfer could be achieved finally.[Methods]The embryos formed by nuclear transfer were put randomly in antioxidant condition,which was different kind and different concentration,undertake the vitro culture to observe its cleavage and blastula rate then analysis the results.[Results]1. In the experiment of effect of NAC on efficiency of Yanbian Yellow Cattle somatic cell clone with different concentration in IVC medium, the result shows that the rate of reconstructed embryo cleavage and blastula development in 0.6mM group was all significantly higher than other groups(P<0.05), so 0.6mM group is the suitable concentration of NAC for Yanbian Yellow Cattle IVC of somatic cell clone as addition alone.2. In the experiment of effect ofβ-ME on efficiency of Yanbian Yellow Cattle somatic cell clone with different concentration in IVC medium,the result shows that the rate of reconstructed embryo cleavage and blastula development in 50μM group was all significantly higher than other groups(P<0.05), so 50uM group is the suitable concentration ofβ-ME for Yanbian Yellow Cattle IVC of somatic cell clone as addition alone.3. In the experiment of effect of EDTA-2Na on efficiency of Yanbian Yellow Cattle somatic cell clone with different concentration in IVC medium,the result shows that the rate of reconstructed embryo cleavage of 50μM group was higher than other groups without statistics differencd but the blastula development rate of 50μM group was all significantly higher than other groups(P<0.05), so 50μM group is the suitable concentration of EDTA-2Na for Yanbian Yellow Cattle IVC of somatic cell clone as addition alone.4. In the experiment of effect of Sodium Citrate on efficiency of Yanbian Yellow Cattle somatic cell clone with different concentration in IVC medium, the result shows that the rate of reconstructed embryo cleavage and blastula development in 0.6mM group was significantly higher than OmM and 0.3mM groups except 1.2mM group(P<0.05), by comprehensive analysis determined that 0.6mM group is the suitable concentration of Sodium Citrate for Yanbian Yellow Cattle IVC of somatic cell clone as addition alone.5. In the experiment of effect of VE on efficiency of Yanbian Yellow Cattle somatic cell clone with different concentration in IVC medium, the result shows that the rate of reconstructed embryo cleavage and blastula development in 100μM group was all significantly higher than other groups(P<0.05), so 100μM group is the suitable concentration of VE for Yanbian Yellow Cattle IVC of somatic cell clone as addition alone.6. In the experiment of effect of EGCG on efficiency of Yanbian Yellow Cattle somatic cell clone with different concentration in IVC medium, the result shows that the rate of reconstructed embryo cleavage in 7.5μM group and 15μM group were significantly higher than 0μM (P<0.05) and blastula development in 15μM group was significantly higher than 0μM and 30μM groups except 7.5μM, by comprehensive analysis determined that 15μM group is the suitable concentration of EGCG for Yanbian Yellow Cattle IVC of somatic cell clone as addition alone.7. In the experiment of effect of Sodium Selenite on efficiency of Yanbian Yellow Cattle somatic cell clone with different concentration in IVC medium, the result shows that the rate of reconstructed embryo cleavage and blastula development in 2.5ng/ml group and 5ng/ml group were all significantly higher than others(P<0.05), but 5ng/ml group had obvious effect than 2.5ng/ml group, so 5ng/ml group is the suitable concentration of Sodium Selenite for Yanbian Yellow Cattle IVC of somatic cell clone as addition alone.[Conclusion]The results shows whatever kind of antioxidant we added, if only under the measurable concentration,all can help conquer the development block resulted by the existence of peroxide,raise the clone efficiency effective indeedly.
