| In this study, hard foxtail-like sophara seed's and Bifidobacterium bifidum were taken as the main experimental material, development a new type green probiotics additives for forage .First of all, experiments carried out on the Oxygen -Resistant Tames of Bifidobacterum bifidum, The acclimated Bifidobacterum bifidum, the living bacterial number was detected by plate culture count method,and identification of bacteria.To make the Foxtail-like Sophara Seed's Dregs-Bifidobacterum bifidum Synbiotics,using the mixed culture method to study influence on the growth of S.pullorum and E.coli in vitro. Screened by the orthogonal test Bifidobacterium bifidum microencapsulation technique encapsulated methods, materials, technology, etc.Finally,the exeperiment studied the cooperation effect of Foxtail-like Sophara Seed's Dregs and microcapsules Bifidobacterum bifidum on immunity of weaned broilers. 350 AA+ chickens being healthy and having even weight were randomly divided into 6 groups, Groupâ… was the control group, Control group fed basal diet(no add Antibiotics of basal diet). The trialâ…¡group was fed basal diet + 0.02% Bifidobacterum bifidum freeze-dried powder, The trialâ…¢group was fed basal diet + 2.0% Foxtail-like Sophara Seed's Dregs, Groupâ…£,â…¤,â…¥was fed basal diet,to witch 0.5%,1.0%,2.0% Synbiotics were added respectively,for 42 days,Every week Thursday the following targets were detected: The index of thymus,the index of spleen,the index of Bursa of Fabricius,the index of appendices tonsil,NDHI Antibody Titer, NK cell activity,IFN-γConcentration and total cholesterol(TC) content of serum. The results showed that:1,After the Bifidobacterum bifidum oxygen-resistant domesticated of live bacteria was 1.5×108cfu / g, species identification is still Bifidobacterium bifidum, and the increase of oxygen-resistant properties.2,The influence of the Foxtail-like Sophara Seed's Dregs-Bifidobacterium synbiotics to S.pullorum ,E.coli with different congcentrations of 0.5%,1.0%,1.5%,2.0% in vitro respectively,The result indicated that Obvious effect bacteriostasic activity of Foxtail-like Sophara Seed's Dregs-Bifidobacterium synbiotics to chicken intestinal pathogenic bacteria. Foxtail-like Sophara Seed's Dregs-Bifidobacterium synbiotics on chicken E.coli and S.pullorum in vitro growth inhibition compared with the control group significant difference (P<0.01), And the proportion with synbiotics increased inhibition also increases.3,The extrusion method was selected for microencapsulating Bifidobacterum bifidum alginate has good bio-compatibility.We had found out the optimum process of making microcapsule and additive magnitude of supporters though the orthogonal experiment.The results showed that the optimum conditions for microencapsulation were established as follows:the alginate concentration as 2.5%,skim milk as 4.0%, calcium chloride as 2.0%,and its livability reached 7.67×107cfu/g. The resistance to stress/bad conditions of Bifidobacterum bifidum microencapsulation including acid resistance and stability during storage had been greatly improved.4,Studies on the effects of the Foxtail-like Sophara Seed's Dregs-Bifidobacterum Synbiotics on immunity function in AA+ Chickens. The results showed that:â‘ On age of 7th, the index of appendices tonsil of 0.5% and 2.0% Synbiotics are higher than control(P<0.05 or P<0.01).â‘¡On age of 14th, the index of appendices tonsil of 0.5% Synbiotics is higher than control(P<0.01);NDHI Antibody Titer of the 0.5% and 2.0% Synbiotics are higher than control(P<0.01); total cholesterol(TC) content of serum of 1.0% Synbiotics is higher than control(P<0.01).â‘¢On age of 21th, the index of Bursa of Fabricius of 1.0% and 2.0% Synbiotics are higher than control(P<0.01); NDHI Antibody Titer of the 1.0% Synbiotics is higher than control(P<0.01); NK cell activity of 2.0% Synbiotics is higher 8.3% than control; total cholesterol(TC) content of serum of 0.5% and 2.0% Synbiotics are higher than control(P<0.05 or P<0.01).â‘£On age of 28th, NDHI Antibody Titer of the 1.0% and 2.0% Synbiotics are higher than control(P<0.05 or P<0.01); total cholesterol(TC) content of serum of 0.5% and 2.0% Synbiotics are higher than control(P<0.05 or P<0.01).⑤On age of 35th, total cholesterol(TC) content of serum of 0.5%,1% and 2% Synbiotics are higher than control(P<0.05 or P<0.01).â‘¥On age of 42th, the index of appendices tonsil of 0.5% Synbiotics is lower than control(P<0.05); NDHI Antibody Titer of the 1.0% Synbiotics is higher than control(P<0.01); IFN-γConcentration the 1.0% Synbiotics is higher than control(P<0.01); total cholesterol(TC) content of serum of 0.5% Synbiotics is lower than control(P<0.01).Conclusion:1,Foxtail-like Sophara Seed's Dregs-Bifidobacterium synbiotics synbiotics significantly increased AA + broiler immune organ index(Particularly the index of Bursa of Fabricius and the index of appendices tonsil). 2,Foxtail-like Sophara Seed's Dregs-Bifidobacterium synbiotics synbiotics significantly increased AA + broiler NDHI Antibody Titer.3,Foxtail-like Sophara Seed's Dregs-Bifidobacterium synbiotics synbiotics can enhance NK cell activity AA + broiler.4,Foxtail-like Sophara Seed's Dregs-Bifidobacterium synbiotics synbiotics can increase serum concentrations of interferon-γ.5,Foxtail-like Sophara Seed's Dregs-Bifidobacterium synbiotics have increased serum total cholesterol function.6,Throughout the results, 1.0% of the Foxtail-like Sophara Seed's Dregs-Bifidobacterium synbiotics can increase the additive amount of AA + broiler immune function, suitable for production practice. |
