PPolysavone was a kind of alfalfa extracts from natural plant alfalfa (Medicago Sativa L) by the water formed after the spray-dried solids alfalfa flavonoids, saponins and polysaccharides was its main active ingredient. The study was conducted to evaluate the effect of Polysavone (extract from alfalfa) on performance, lipid of tissue and egg in early (28~35 wk) and later (58~65 wk) period of Hy-line brown laying hens and the aim of this study was to provide a theoretical basis for the apply of Polysavone in laying hens.Research method:The study was divided into two stages.384 Hy-line brown laying hens were separately selected from 28 wk and 58 wk laying hens, and were randomly assigned to 4 groups, each treatmen included 6 replicates,16 layers per replicate. Ttreament I was the control group fed with basal diet, treatmentⅡ,Ⅲ,Ⅳfed with the basal diet supplemented with Polysavone 500 mg/kg,1000 mg/kg and 1500mg/kg to investigate the effects on performance, cholesterol and triglyceride concentration in yolk and egg, cholesterol and triglyceride concentration in liver, cholesterol in chest muscle, serum lipid,and lipoprotein lipase activity in serum.Exp. One Effect of Polysavone on performance of laying hensThe feeding trial lasted for 49 days.The laying rate, feed-egg ratio, average daily feed intake and average egg weight were caculated weekly. The results showed that the three treatments of Polysavone had no significant effect on performence of 28 wk laying hens (P>0.05).The three-dosage Polysavone groups all significantly decreased feed-egg ratio (P<0.05), and the group of 500 mg/kg and 1000 mg/kg Polysavone significantly increased laying rate of 58 wk laying hens (P<0.05).The three-dosage Polysavone groups had no effect on average daily feed intake and average egg weight(P >0.05).Exp. Two Effects of Polysavone on performance, lipid of egg and tissues in laying hensEgg samples were randomly sampled once 12 day from the first day of experiment period, taking an egg each repetition. Cholesterol and triglyceride levels in egg were detected. A chicken was randomly selected at 2,4,6,7 wk of experiment period from each reptition of each treatment, and fasted blood 5 mL, from heart, put iaside for 30 minutes at room temperature, centrifugated 10 minutes at 3000 revolutions per minute. Serum was separated and serum lipids were detected. After chicken was slaughtered, the abdominal cavity was immediately opened. Then the same part of the liver was removed, with normal saline washing away blood, sub-installed in 5 mL centrifuge tube. Cholesterol and triglyceride of liver were detected. The same part of the chest muscle was removed.The chest muscle was shredded, placed in petri dishes, and placed in 65°C oven after weighing, and drying to constant weight, weighed, grinded.Then air-dried sample was preparated. Cholesterol in chest muscle was detected. The results showed that compared with the control group, dietary Polysavone 1000 mg/kg had significantly decreased cholesterol and triglyceride concentration in yolk of 28 wk laying hens(P< 0.05), and the group of 1000 mg/kg,1500 mg/kg Polysavone significantly decreased cholesterol and triglyceride levels in egg (P< 0.05). Compared with the control group, dietary Polysavone 1000 mg/kg had significantly decreased triglyceride concentration in yolk and cholesterol in egg of 58 wk laying hens (P< 0.05), and dietary Polysavone 500 mg/kg,1000 mg/kg and 1500 mg/kg all significantly decreased triglyceride levels in egg (P< 0.05), and had no significant effect on cholesterol in yolk (P> 0.05). Three treatments of Polysavone all significantly decreased cholesterol concentration in liver of 28 wk laying hens(P<0.05).the group of Polysavone 1000 mg/kg and 1500 mg/kg significantly decreased triglyceride concentration in liver of 28 wk laying hens(P< 0.05).Compared with the control group, dietary Polysavone 500 mg/kg,1000 mg/kg and 1500 mg/kg significantly decreased cholesterol and triglyceride concentration in liver of 58 wk laying hens(P<0.05).Three treatments of Polysavone all significantly decreased cholesterol concentration in chest muscle of 28 wk laying hens(P<0.05), but had no significant effect on cholesterol concentration in chest muscle of 58 wk laying hens(P>0.05).Three treatments of Polysavone had no significant effect on cholesterol, triglyceride, high density lipoprotein cholesterol, low density lipoprotein cholesterol in serum of 28 wk and 58wk laying hens (P>0.05).Compared with the control group, dietary Polysavone 1500 mg/kg had significantly increasd very low density lipoprotein cholesterol of 58wk laying hens (P<0.05),but had no effect on 28 wk laying hens. Dietary Polysavone 1000 mg/kg significantly inceased lipoprotein lipase activity in serum of 28 wk laying hens(P<0.05).Dietary Polysavone 1500 mg/kg had significant effect on inceased lipoprotein lipase activity in serum of 58 wk laying hens(P>0.05).
