Pudong chicken was an excellent native breed in China. In this study, 15 microsatellite DNA markers were selected to analysis the allele frequency, heterozygosity(H), mean heterozygosity, polymorphism information content(PIC) and genetic relationships among Pudong chicken and other 6 native chicken breeds, the relationship between 15 microsatellite loci and body measurements, body weight, slaughter traits was also analyzed. The results were as follows:1. The polymorphisms of 15 microsatellite loci detected in 7 Chinese indigenous chicken breeds were abundant and a total of 75 alleles were observed. The mean number of alleles was 5(2-8) in every microsatellite locus; Mean heterozygosities of the 7 Chinese indigenous chicken breeds varied from 0.4825 to 0.6189,the PIC ranged from 0.4943 to 0.6153, and the mean effective number of alleles in 29 microsatellite loci ranged from 2.5 to 3.2; Pudong chicken contented the highest H,PIC and Guifei chicken contented the lowest;2. The result of genetic diversity indicated that Pudong chicken was significant difference with other 6 breeds(0.1872~0.6003). The seven chicken populations were clustered into two groups based on the dendrogram on the basis of the Nei's genetic distance. The first big group including 6 Chinese indigenous chicken breeds was divided into two groups as follow: Liyang chicken and Shiqiza chicken clustered together first, followed by Anka chicken and Pudong chicken, formed the first group; the second group was made up of two indigenous chicken breeds: Beijing Fatty chicken and Hailan chicken . The second big group comprised Guifei chicken respectively. As a whole, the clustering diagram reflected well the phylogenitic evolutionary and breeding history of those chicken breeds.3. The relationship between microsatellite markers and thirteen dressing characters showed that MCW0014 and MCW0295 were remarkable relationship with chest depth;ADL0278 and MCW67 were remarkable relationship with the chest bone length;ADL0146 was remarkable relationship with the pelvis width;LEI094,MCW0014 and MCW67 were remarkable relationship with the leg muscle rate;the relationship between MCW0094 and the body weight was extremely significant.The different genotypes of the same microsatellite DNA marker which linkage with traits were compared by way of Duocan, the conclusion was as follows: AC of MCW0014 was remarkable correlative to the chest depth(P<0.05);AA of ADL0146 was remarkable correlative to the thurl width(P<0.05).
