The genetic diversity of 211 individuals of four local sheep in Henan province was studied using 27 microsatellite locis(including Small tail sheep,Large tail sheep,Yuxi fat-tail sheep and Taihang fur sheep), Numbers of alleles, polymorphism information contents, heterozygosity, HW Test, private alleles, dominant alleles, coefficient gene differentiation,standard genetic distance and Nei geometrical genetic distance among the four sheep populations were computed with and dendrogram were constructed to evaluate the genetic variability within breeds and genetic relationship among breeds. The study was aimed to provide scientific basis for proper development and protection of these sheep breeds. The results were as follows :1. 27 microsatellites loci were used to evaluate the genetic polymorphisms in four sheep breeds, the 27 loci were polymorphic and 370 alleles were detected, with a mean of 13.70, which indicated that the 27 loci were morepolymorphic in the four sheep breeds.2. The allele frequency was got with the POPGENE program. Accordingly,we calculate PIC,He ,and Ne,This showed that there were great variability and broad genetic base in the four sheep breeds.3.Some dominate alelles and private alelles were shown to be important in the costruction of the population structure.4. Heterozygosity in subpopulation, total heterozygosity, and coefficient gene differentiation of the 27 microsatellites loci were calculated, We find the genetic which demonstrated small genetic differentiation among the four sheep breeds.5. Genetic distance of the Ds and Da was calculated among the four sheep breeds by DISPAN program. The results show that the genetic distance between populations vary inapparently. According to DS and Dcoa,We make NJ and UPGMA cluster.The reconstruct tree by NJ method match with that of by UPGMA method,so the result is consistent .6.The results of Chis which Hardy-Weinberg equilibrium in showed that all sheep breeds do not fit these 27 locus or selection, genetic drift etc.
