As an ideal veterinary drugs and feed additives, Olaquindox is antibacterial, protein assimilating, growth promoting. But the improper use of olaquindox, lead poisoning cases reported common, in March 2009 by the CCTV "Weekly Quality Report" reported that occurred in the city of Lianyungang in Jiangsu Province for carp aquaculture feed containing "olaquindox" caused the mass mortality of carp. Driven by the economic interests, the large doses of abuse result in residues in animal products, caused great harm.According to GB15193.1-2003 and toxicological analysis of food security,In this experiment, we use Kunming (KM) mice as animal models and exposured by gavage way, systematically studied the acute and accumulation toxic characteristics of Olaquindox. We also further studied the growth performance of mice, physiological functions of organs and tissues, the pathological injury and blood parameters.In acute toxicity test, modified Kou's method was used to test oral Olaquindox LD50 of KM mice. The KM mice were randomly divided into 5 groups, fed with equal volume of unequal concentration Olaquindox, observed clinical symptoms, measured organ and blood biochemical index and pathological observation.(1) For Olaquindox,the median lethal dose of the KM mice (LD 50) was 2668mg/kg,95% confidence interval range was 2291-3106mg/kg, the results showed that the drug is of low toxicity substances. (2)The higher the dose, the earlier the onset of symptoms and the more serious, it showed a dose-effect relationship, the high dose of Olaquindox can have a direct lethal effect on mice.(3) Organ index determination, pathological examination and histological sections showed that liver and heart are Olaquindox target organ of acute poisoning, acute cardiac damage in mice will lead to cardiac and respiratory failure occurs. (4) The blood biochemical parameters showed that, groupâ… andâ…¡in the serum total protein, albumin, A/G ratio both lower than the control control group,and for the tow test groups of mice,the index of serum reduced with the dose increase. groupâ… andâ…¡in the aspartate aminotransferase, lactate dehydrogenase, alkaline phosphatase activity are both higher than the control group, and for the tow test groups of mice,the index of serum increased with the dose increase.Incremental dose toxicity method was used to do accumulation toxic test,40 healthy mice were randomly divided into 2 groups, experimental group contained 30 and control group 10, housed separately. the initial dose for test group was 1/10 of LD 50, daily exposure,four days as a period, and increase 1.5 times each period, the feeding continued until half of the death. During the experiment, the growth performance of mice and the clinical symptoms was observed, the dead mice were dissected for organ histopathology observation, calculating the viscera indices and measuring blood biochemical parameters.(1) Olaquindox cumulative coefficient in mice is 3.69, it's in the range of 3 to 5, with moderate cumulative toxicity. (2)The experiment also found that different levels of additive olaquindox in mice do not affect the growth of the same; in lower dose level less than 600.30 mg/kg, Olaquindox significantly affected the growth in mice, without toxic symptoms, nor death. But as the dose increases, when more than 900.45 mg/kg, the weight gain slowed or negative growth, and showed symptoms of poisoning, began to die and reached the death peak.(3) The most serious pathological damage to mice were liver and kidney damage, heart, spleen, lung, stomach and bowels also are subject to varying degrees of damage. (4) In poisoned mice, total protein, albumin, A/G of serum content decreased, ALT, aspartate aminotransferase, lactate dehydrogenase activity increased and urea nitrogen and creatinine levels also increased, indicating that, olaquindox has significant damage in mice liver and kidney.Therefore, in the actual livestock production, although Olaquindox can promote the animal's growth rapidly to gain more economic benefits, but due to the biological toxic hazards, and domestic unreasonable abuse is widespread, not only seriously affected the quality and exports of livestock and poultry product in China, but also was a great potential threat to the health of consumers.
