It is still a puzzle for repairing peripheral nerve defects. Nerve autograft is still the best choice. But it is limited by the resource and hyposensitivity. Seeking a perfect graft which could be applied to clinic has always been a hotpoint. With the tissue-engineering developing, we can look for new ways for peripheral nerves repairing. Objectives: SC cultured from newborn Sprague-Dawley rats were planted on PLA membranes, and the sciatic nerve defect was repaired by PLA tubes planted with SC to research SC growth in PLA membranes and the capability that activity SC surviving and growing in PLA tubes in vivo. The possibility of repairing peripheral nerves with the tissue-engineering artificial nerve was studied, offering foundation for clinical application. Method: The sc suspension, isolated from sciatic nerves and brachial plexus of 40 newborn SD rats, and regulated to 106 cells/mi, were planted on the PLA membrane, 96 wells cultural plate and 20m1 cultural bottle. Then the growth of SC was examined under phase contrast microscope and scanning electron microscope at 1st, 3rd, 5th, 7th, 14th day after culture. After being cultured three days in the culture bottles, SC were planted in PLA tubes with concentration of 1 o~ cells/mi. The complex were cultured for another 24h, and then used to repair sciatic nerve defects in 10 adult SD rats. Middle segment of nerve grafts were excised for light microscopy and transmission electron microscopy at 4, 8,12 weeks. Resultes: 1. SC can grow on the PLA membrane. 2. SC can grow in the PLA tube. The regeneration axons form myelin sheath. 3. Accidentally, striated muscle cells have been found in regeneration nerve fibres. Conclusions: 1. SC is compatible with the PLA membrane. 2. After the PLA tube is planted with SC, SC can grow in, and the regeneration nerve fibres form. 3. As peripheral nerve tissue-engineering seed cells, SC must be studied for more details.
