Insulin-like growth factor (IGF) is a kind of multi-function cellproliferation regulation factor, synthesized in a variety of tissue. IGFs can exert not only endocrine function but also mitogenic effects within the local tissue environment through autocrine and/or paracrine mechanisms. The IGF system is expressed in human uterus, is believed to involve in proliferation and differentiation of the uterine endometrium during the menstrual cycle, enhancement trophoblast invasion , promotion placental development ,embryonic and fetal growth in human pregnancy. But the accurate role mechanism of IGF system in human uterus is still not very clear. Some expresses of IGF system have still dispute. Now, there is no research report about the expression of IGF- II in human villus and decidua during early pregnancy in China and foreign document information is very few too. We employed the SP immunohistochemistry techniques with 6 l~f?~tt~z~ monoclonal antibodies specific to IGF- II -. ER~ PR to locate the IGF- IL ER and PR in human villus and decidua during early pregnancy ,we also used the histology score for semiquantitative analysis. Correlation analysis among the expression of IGF- IL ER and PR were carried out too. The purpose of our study was to offer some theory evidences for raising the successful rate of IVF-ET by discuss the mechanism of IGF- II regulate trophoblast invasion , promote embryonic and fetal growth in human pregnancy. Materials and Methods We selected 30 pregnant women between Days 40 to 60 of intrauterine gestation who are not more than 35 years old and came to terminate pregnancy by induced abortion voluntarily in Zhejiang Obstetrics and Gynecology hospital from December 2000 to January 200 1. Methods 1.materials: We got fresh villus and deciduas tissue by clean negative pressure inhale plastic tube, then rubed bloodstain with dry gauze, froze materials in liquid nitrogen and store at -80 ~C ultralow temperature refrigerator immediately. 2.methods: We used the SP immunohistochemistry techniques with monoclonal antibodies specific to IGF- II ER. PR to study the expression of IGF- H ER and PR in human villus and decidua during early pregnancy. 7 3. Result judge standard: We observed the SP dyeing sections under the CHK model olympus microscope of 400 times. we selected 5 field of visions in cytotrophoblast, syncytiotrophoblast or decidual stromal cell dense district, then counted 100 same kind cells on every field of vision, Read out respectively the number of negative, weak positive, positive and strong positive cells, then we had semiquantitative analysis with the method of histology score. The Calculate formula is H-score= Z pi (i+1) 4.Statistical analysis: All data was processed with SPSS 10.0 software bag for windows. Single factor analysis of variance and Pearson correlation analysis were used for statistical evaluation. The level of significance was selected as a =0.05, 2-tailed.
