Objective To assess ischemia and reperfusion injury in partial hepatectomy in rabbit model after continuous or intermittent vascular inflow occlusion, and to appraise the protection of ischemia preconditioning for the liver after vascular occlusion. Methods 48 rabbits were divided into four groups randomly. In group A (control group, n=12), simple laparotomy was performed only. In group B (continuous vascular inflow occlusion group, n=12), the vascular inflow was occluded for 40mm. In group C (intermittent vascular inflow occlusion group, n=12), the vascular inflow was occluded for 20mm, After opened for 10mm, the inflow was occluded for 20mm again. In group D (ischemia preconditioning group, n12), after the inflow was occluded and opened for 10mm respectively, the vascular inflow occlusion was performed for 30mm. During the period of vascular occlusion (within the last 10 minutes of ischemia), left partial hepatectomy (about 35% of total liver) was carried out in group B, C and D. Liver function including asparate aminotransferase (AST), alanine aminotransferase (ALT), Lactate dchyolro genase (LDH), and activitives of superoxide dismutase (SOD) in plasma were detected to analysis the liver injury. The lever of adenine 1-5- triphosphate (ATP) in liver tissue was examined by the velocity of respiratory chain in mitochondrial. The liver biopsy was used to evaluate the degree of liver damage by light microscope. Results ~D Compared with group A, the liver function including AST, ALT, and LDH after occlusion for 40 minutes, reperfusion 60 minutes, 24 hours and three days after hepatectomy was significantly higher (P<0.05). Lower levers of AST, ALT and LDH were found in group D than that in group B and C (P<0.05). ?The levers of SOD in group B, C and D were significantly lower than that in group A (NO.05). However, the lowest lever of SOD was present in group D after 40minutes 4 occlussion and 6Ominutes reperfusion than that in group B and C (P
