Gastric carcinoma and colorectal carcinoma are common disease. Their mortalities are very high. Basical and clinical research on gastric carcinoma and colorectal carcinoma is always of importance. The nm23 gene has been heralded as a metastasis suppressor gene. Two highly homogenous genes, nm23-Hl and nm23-H2, have been described, both located on the long arm of chromosome 17, coding for the 18.5 and 17 KD proteins respectively. The nm23-Hl and nm23-H2 gene products have been shown to be identical to human nucleoside diphosphate (NDP) kinased A and B. In a number of studies of human malignant tumors, such as breast carcinomas, malignant melanomas and hepatocellular carcinomas, an associantion between reduced expression of nm23-Hl mRNA or its protein and an increasing metastatic ability could be demonstrated. However, ther are still several studies reporting a lack of correlation between nm23 expression and metastatic potential in gastric carcinoma and colorectal carcinoma. In the current study, the exprssion of nm23-Hl was examined in a series of gastric carcinoma and colorectal carcinoma.Materials and Methods41 gastric carcinoma tissues and normal gastric mucosa were obtained from patients operated on in the First Affiliated Hospital of Zhejiang University from August 2001 to February 2002 , 28 males,!3 females, mean 57.5 years old (age ranged from 29 to 80). 76 colorectal carcinoma tissues, para-carcinoma tissues and normal colorectal mucosa were also obtained form patients operated on in the same hospital from March 2001 to November 2001, 46 males, 30 females, mean 58.9 years old (age ranged from 22 to 87). The expression of nm23-Hl was analyzed by reverse transcriptase-polymerase chain reaction (RT-PCR), immunohistochemistry (IHC) and flow cytometry (FCM). And the correlation of the expression of nm23-Hl with different histomorphologic variables was evaluated.ResultsNo significant difference of expression of nm23-Hl mRNA or protein was detected between gastric carcinoma and normal gastric mucosa. The expression of nm23-Hl protein in gastric carcinoma was correlated with histological classification. Higher level expression was detected in adenocarcinoma, mucmous adenocarcinoma than signet ring carcinoma, undifferentiated carcinoma (x2=12.097,p<0.01). No significant relations were found between the expression of nm23-Hl mRNA or protein and the sex, tumor size, pT category, metastasis or TNM category.Higher level expression of nm23-Hl mRNA in colorectal carcinoma was detected in carcinoma tissue than normal colorectal mucosa (p<0.05). Expression of nm23-Hl protein detected by IHC, in carcinoma tissue was higher than para-carcinoma tissue (p<0.05) and normal colorectal mucosa (p<0.001). However,detected by FCM, expression of nm23-Hl protein in carcinoma tissue was lower than normal colorectal mucosa (p<0.05). The expression of nm23-Hl mRNA in colorectal carcinoma was correlated histological classification. Higher level expression was detected in signet ring carcinoma than papillary, tubular and mucinous adenocarcinoma. (x2=8.825, p<0.05). No significant relations were found between the expression of nm23-Hl mRNA or protein and the sex, age, pT category, metastasis or Dukes category.No significant coincidence was found between expression of nm23-Hl mRNA and nm23-Hl protein in gastric carcinoma as well as colorectal carcinoma.Conclusions1. The expression of nm23-Hl gene in gastric or colorectal normal mucosa can be detected; expression of nm23-Hl mRNA and protein is higher than normal colorectal mucosa, while expression of nm23-Hl protein in the cell membrane in colorectal carcinoma is lower than normal colorectal mucosa.2. The expression of nm23-Hl in gastric carcinoma and colorectal carcinoma is correlated with histological classification.3. The expression of nm23-Hl mRNA and its protein in gastric carcinoma or colorectal carcinoma is lack of coincidence.
