Objective: To investigate the relationship of HLA-DQA1 gene alleles and type 2 diabetes mellitus ( T2DM ) in Guangxi Chinese Zhuang nationality. Methods: Case-control study, the classifications of HLA-DQA1 alleles between T2DM group (n = 48) and normal controls (n = 49) in Guangxi Chinese Zhuang individuals were identified by Polymerase Chain Reaction-Sequence Specific Primers(PCR-SSP) technology. The level of the insulin and c-peptide at the point of fast plasma and postprandial 2 hours were measured with Radioimmunoassay (RIA) technique. Results: The frequencies of DQA1*0401, DQA1*0301 in T2DM group were significantly higher than those in the control group (P <0.05, relative risk (RR) = 4.559 and RR = 10.379, respectively ) .The frequency of DQA1*0601 occurred in the T2DM group were remarkably decreased than that in the controls (P<0.01, RR = 0.134); compared with patients of negative alleles, the level of the c-peptide of fast plasma (Oh) and postprandial 2 hours (2h) were significantly lower in T2DM patients with HLA-DQA1*0301 + genotype as compared with T2DM patients with HLA-DQA1*0301" genotype (P<0.01); the frequency of DQA1*0301 in the group of Oh-CP^0.3nmol/L and 2h-CP^0.6nmol/L were significantly increased than the group of Oh-CP>0.3nmol/L and 2h-CP>0.6nmol/L (P<0.01, RR= 4.923 , 5.506 respectively). Conclusions:The results suggested that allele gene DQA1*0401, DQA1*0301 may contribute to the susceptible alleles of T2DM in Guangxi Chinese Zhuang nationality, DQA1*0601 may be importantly protective alleles; DQA1*0301 genotype could predicate that beta cell function would be more easily exhausted in T2DM patients.
