Objective To investigate the expression of P27KIP1, P21WAF1, CyclinE, Ki-67 in transitional bladder cancer, the relationship of P27KIP1, P21WAF1, Cyclin E ^ Ki-67 in transitional bladder cancer.Methods 80 transitional bladder cancer specimens and 15 normal bladder specimens were examined for P27KIP1, P21WAF1, Cyclin E by in situ hybridization and immunohistochemistry.Results The positive of P27KIP1, P21WAF1, Cyclin E ^ Ki-67 for transitional bladder cancer was 80% (64/80); 87.5 % (70/80) ; 75% (60/80); 80% (64/80 )0 To investigate the relationship among P27KIP1, P21WAF1, CyclinE, Ki-67 and clinical pathology in transitional bladder cancer by in situ hybridization and immunohistochemistry. Low protein levels of P27KIPl , low protein levels of P21WAF1 were associated with low staining of Cyclin E. Low staining of Cyclin E and high Ki-67 index correlated with poorly differentiated grade, muscle invasion and lymph node metastsis. Cyclin E protein levels was inversely related with Ki-67 index. Kaplan-Meier plots of survival rate in patients with low versus high P27 staining showed that low protein levels of p27 were associated with a shortened disease-free and overall survival. Onmultivanate analysis using Cox proportional hazards model, low protein levels of P27and high Ki-67 index were independent predicators of shortened disease-free, and lowprotein levels of p27, low staining of Cyclin E and high Ki-67 index of overallsurvival.Conclusions It is suggested that decreased protein levels of P27Kn\ P21WAF1,cyclin E> Ki-67 play a role in the progression of bladder cancer and to evaluate theseprotein levels may be useful in management of the diseases.
