Objective The polypeptides (PCF,Mr:800-1000) was isolated from chlamys far-reri using enzyme engineering technica. We established the moder of middle cerebral artery occlusion (MCAO) in the rats to study the protection of PCF on brain nevers after cerebral ischemia and reperfusion and its mechanism.Methods 64 Wister rats were randomly divided into four groups; sham group and MCAO model groups which were randomly divided into PCF group, distilled water group and control group. Neurons in the cerebral ischemia penumbra were observed dyeing by Cresyl Violet; the activities of SOD, GSH-Px and the amounts of MDA in the ischemia penumbra were determined by Zymchemistry; the expression of Bcl-2 gene and Bax gene in the ischemia penumbra were examined by immunochemical technical.Results The results of Cresyl Violet dye indicated that neurons in parietal cortex of PCF group were less damaged than that of control group. Comper to control group, the activities of SOD in PCF group were enhance from 74. 65+17. 18 Nu/ml to 120. 37?20. 35 Nu/ml? the activity of GSH-Px from 21. 29?. 07U to 29. 38?. 15U,the a-mounts of MDA were decrease from 79. 65 + 15. 62 to 39. 56?3. 19. The results indicated that PCF could enhance the activities of SOD, GSH-Px and might decrease the a-mounts of MDA in brain tissue. The PCF also could raise the expression of Bcl-2 gene and inhibit the expression of Bax gene in the ischemia penumbra. The observed indications of the PCF groups are significantly different compared with control group (P < 0.01).Conclusion PCF has the protective effects on brain nerves after cerebral ischemia and reperfusion. Its mechanisms are its abilities of raising the expression of Bcl-2 gene and inhibiting the expression of Bax gene in the ischemia penumbra, increasing anti-oxi-dative enzymes in brain tissue and inhibiting the lipid to be peroxided.
