OBJECTIVE: (l)To explore popularizingGDI05 as a kind of basic marker of active vascular endotheliumby comparing the expression of CD105,an active vascularendothelial marker and pan-vascular endothelial marker FactorVIII in colorectal carcinoma.(2)To explore the expression of VEGF and CD105 in colorectalcarcinoma and their relationship with clinicopathologic factorsso to provide experimental evidence of curing colorectalcarcinoma by blocking VEGF and CD105 .METHODS (1)44 cases of carcinoma specimens were collected,with 20 cases of colorectal tissue regarded as the controlgroup.The S-P immunohistochemical tecnique was used to detectthe expression of CD105 and Factor VIII in both theexperimental and control group .(2) The same groups as above were used. The S-Pimmunohistochemical tecnique was adopted to detect theexpression of VEGF in the two groups .RESULTS (l)The MVD value of the 44 carcinoma marked byCD105 was 144.98 ± 32.67/100 X (scope);higher than that byFactor VIII which was!00.39± 18.70/100X (scope)(P<0.01);bothwere significantly higher than those of control tissue which were41.23 ± 11.64/100 X (scope)and 45.70 ± 15.18/100 X(scope)(P<0.01).(2) The positive rate of VEGF of the 44 carcinoma was 59.1%(26/44),significantly higher than that of control tissue[15%(3/20)](P<0. 01). Both the expression of VEGF and CD105(MVD) were positively related with depth of invasion -. lymph-node metastasis and Dukes stage(P<0.01 or P<0.05);but their relations to histological grade were notsignificant(P>0.05). There was a significant correlation between the expression of VEGF and CDIOS(MVD) [ri=0,720 ,P<0.01] higher than that between VEGF and FACTOR VIII(MVD) [r2=0.432 ,P<0.05].Further analysis proved that ri>F2 [P<0.05].CONCLUSION (1)CD105 is superior to Factor VIII in marking MVD of colorectal carcinoma and is fit to be taken as a formal active vascular endothelial marker in this tissue. (2) The expression of VEGF and CDIOS(MVD) are positively related with the clinicopathologic state of lymph-node metasstasis, depth of invasion and Dukes stage in colorectal carcinoma .There is a positive correlation between the expression of VEGF and CDIOS(MVD) in this tissue. It might be helpful to the treatment of colorectal carcinoma by simultanuously inhibiting VEGF and CD105.
